Quality assessment of platelet concentrates prepared by platelet rich plasma-platelet concentrate, buffy coat poor-platelet concentrate (BC-PC) and apheresis-PC methods

Abstract: Background:Platelet rich plasma-platelet concentrate (PRP-PC), buffy coat poor-platelet concentrate (BC-PC), and apheresis-PC were prepared and their quality parameters were assessed.Study Design:In this study, the following platelet products were prepared: from random donor platelets (i) platelet rich plasma - platelet concentrate (PRP-PC), and (ii) buffy coat poor-platelet concentrate (BC-PC) and (iii) single donor platelets (apheresis-PC) by different methods. Their quality was assessed using the following … Show more

“…Despite the fact that the platelet number may be responsible for these observed variations, a large study reported a poor correlation between platelet counts and specific growth factor concentrations. 18,44 However, other studies demonstrated a clear effect between the growth factor content of platelets and the biological activity that was induced. 45 Further, it is well known that besides the growth factors, platelets contain a variety of other proteins or chemokines, which also might influence the reaction of cells or tissues.…”

“…[456] New trends in the preparation of platelets showed a shift toward the use of the buffy coat method. It has been suggested that this method causes less platelet activation and damage during platelet preparation.…”

“…Platelet quality can be evaluated in vitro using several parameters such as visual appearance, metabolic activity, volume, platelet count, and WBC count per unit. After transfusion, platelet quality can be evaluated using corrected count increment (CCI) and percentage platelet recovery (PPR) [27,54].…”

“…Singh et al [54] found that apheresis-platelet concentrate units from the CS3,000+ instrument showed better swirling and platelet counts than platelet rich plasma-platelet concentrates; however, Mintz et al [37] confirmed that spherical platelets may revert to the discoid form, and that non-discoid platelets may retain suitable functionality post-transfusion.…”
