Quantification of abscisic acid in grapevine leaf (Vitis vinifera) by isotope-dilution liquid chromatography–mass spectrometry

Abstract: A specific, sensitive, precise, and accurate method for the determination of abscisic acid (ABA) in grapevine leaf tissues is described. The method employs high-performance liquid chromatography and electrospray ionization-mass spectrometry (LC-ESI-MS) in selected ion monitoring mode (SIM) to analyze ABA using a stable isotope-labeled ABA as an internal standard. Absolute recoveries ranged from 72% to 79% using methanol/water pH 5.5 (50:50 v/v) as an extraction solvent. The best efficiency was obtained when th… Show more

“…ABA-glucosides exhibit little or no biological activity, but appear to be a transported form of ABA [40]. In general, leaves have a higher ABA content than roots, and developing seeds and ripening fruits are usually rich sources of ABA [41].…”

“…Tandem analysis with mass spectrometry is widely used to identify ABA and its metabolites, with one (Turecková et al, 2009;Zhou et al, 2003;Ross et al, 2004;LopezCarbonell et al, 2009) or at most two transitions (Gómez-Cadenas et al, 2002;Vilaró et al, 2006) for each compound. However, even with different retention times, the use of only one MRM transition for each analyte may also be problematic.…”

“…The use of mass spectrometry (MS) coupled with an appropriate separation technique such as high-performance liquid chromatography (HPLC) (Ross et al 2004, Vilaro et al 2006, Hou et al 2008, L ó pez-Carbonell et al 2009 ) or gas chromatography (GC) (Cvikrov á et al 1998, Kamboj et al 1999, M ü ller et al 2002, Okamoto et al 2009 ) allows the determination of trace amounts of ABA down to nanograms/ gram fresh weight (ng/g fw). However, most of them require very expensive instruments and complicated sample treating.…”
