Quantification of human angiotensinogen by a novel sandwich ELISA

Abstract: The urinary angiotensinogen excretion rates show a clear relationship to kidney angiotensin II content, suggesting that urinary angiotensinogen may serve as an index of angiotensin II-dependent hypertensive rats. However, simple and accurate methods to measure human angiotensinogen are unavailable at this time. We have developed two antibodies and a sensitive and specific quantification ELISA system for human angiotensinogen to be applicable to human subjects. The ELISA is able to detect human angiotensinogen … Show more

“…Recently, an ELISA that used antiangiotensinogen antibody was reported (37,38). In contrast to our measurement that detects intact angiotensinogen only, the ELISA detects intact angiotensinogen as well as its inactive residue, the des-angiotensin I-angiotensinogen (des-AngI-Ao), cleaved by renin (39).…”

“…Above 2 studies used indirect measurements to assess the urinary angiotensinogen levels because simple and accurate methods to measure human angiotensinogen directly are unavailable at this time. Recently, two independent groups have developed enzyme linked immunosorbent assay system to measure angiotensinogen directly [26,54]. Outcomes of clinical studies using the direct measurement of human angiotensinogen are expected in the near future.…”

“…The primary antibody against human angiotensinogen was raised by Zymed as a custom service and was characterized previously [26] and the concentration for immunohistochemistry was 1:6,000.…”
