Quantification of Pathways of Glucose Utilization and Balance of Energy Metabolism of Rabbit Reticulocytes

Abstract: In this work it is demonstrated that glucose constitutes the main substrate of energy metabolism of rabbit reticulocytes under aerobic conditions in the presence of 5 mM glucose. Amino acids and fatty acids are minor sources of energy. The shares of processes utilizing glucose in reticulocytes were estimated from tracer experiments. A new mathematical technique used permits the derivation of closed terms for the specific radioactivity of single positions of C atoms of the metabolites of the citrate cycle. By m… Show more

“…present in plasma samples. We also identified the metabolite 2,3-diphosphoglycerate that is generated in the Rapoport Leubering cycle during erythrocyte glycolysis [32]. Although hemolysis introduces metabolites of erythrocytes, this did not affect our comparisons of AIBP knockout to control samples because the erythrocyte derived 2,3-diphosphoglycerate was…”

“…Since only 25% of this ATP is in the metabolic pool, we obtain the utilization rate of 0.33 nmol of ATP/min/106 cells or a turnover time of 9.1 min. In comparison to other cells types Siems et al (1982) report that rabbit reticulocytes consume about 200 mmol of ATP/h/L of cells at 37°C and that erythrocytes consume 2 mmol of ATP/h/L. Verhoeven et al (1984) report that platelets consume ATP at a rate 2.5 times higher than rabbit reticulocytes but about at the same rate as human neutrophils and lymphocytes.…”
