Quantitative Analysis of Interleukin‐1‐Alpha Gene Expression in Middle Ear Cholesteatoma

Abstract: Regardless of its origin, cholesteatoma is characterized by the presence of a keratinizing epithelium with an hyperproliferative behavior leading to a very important bone resorption. Previous studies have demonstrated overexpression of interleukin-1 (IL-1 protein in middle ear cholesteatoma by immunohistochemistry and enzyme-linked immunosorbent assay, suggesting a significant role for IL-1-alpha. In this study, the presence of IL-1-alpha messenger ribonucleic acid (mRNA) was quantified by in situ hybridizatio… Show more

“…Several previous studies examined the role of inflammatory cytokines in the pathogenesis of cholesteatoma, including the osteoclastogenic tumor necrosis factor-alpha (TNF-␣; 6,8,19,22,23). Serum levels of TNF-␣ were higher in patients who had more than 2 ossicles destroyed (22).…”

“…A number of studies [15,16] have attempted to delineate a Th1/Th2 paradigm of inflammatory events in cholesteatomas, in which Th1 cells characterized by the production of interferon-␥ , TNF-␣ and IL-1 are likely to be dominant. A third T cell subset known as Th17 was identified and characterized by the production of IL-17 [17,18] .…”

“…Marenda & Aufdemorte 8 and Bujia et al 10 described Interleukin 1 (IL1) in acquired cholesteatoma. They agreed in their findings reporting IL 1 immunoexpression in all layers of the epithelium and subepithelium of the cholesteatoma.…”

“…High concentrations of IL-6 were related to erosion of ossicular chain and presence of intra-operative granulation tissue. IL-8 promotes the production of collagenases, which are proteins involved in process of bone lysis 8,10,11 . In reference to immune localization Tumor Necrosis Factor-Alpha (TNF-α) in acquired cholesteatoma, the authors disagreed.…”
