Quantitative Bead-Based Flow Cytometry for Assaying Rab7 GTPase Interaction with the Rab-Interacting Lysosomal Protein (RILP) Effector Protein

Abstract: Rab7 facilitates vesicular transport and delivery from early endosomes to late endosomes as well as from late endosomes to lysosomes. The role of Rab7 in vesicular transport is dependent on its interactions with effector proteins, among them Rab-interacting lysosomal protein (RILP), which aids in the recruitment of active Rab7 (GTP-bound) onto dynein–dynactin motor complexes to facilitate late endosomal transport on the cytoskeleton. Here we detail a novel bead-based flow cytometry assay to measure Rab7 intera… Show more

“…Iem‐spFRET combined with E‐FRET not only shown reagents‐induced interaction between A 1 R and A 2A R changes of whole cell but also visualized partial differences within same cell. Research shows that interactions between proteins often mediate biological mechanisms [57] (e.g., the transport of receptors [58, 59], regulating the metabolites [60], lipid diffusion [61], pro‐inflammatory responses [62]), and may become a potential therapeutic target [63–65]. Therefore, our results on the spatio‐temporal study of A 1 R and A 2A R interaction may contribute to reveal many clues on the function and regulation of A 1 R‐A 2A R heterodimer.…”

“…Rabs have also been implicated in cancer progression, with the consequence that targeted Rab inhibitors are under development (Salehi et al, 2009; Hong et al, 2010; Agola et al, 2015; Raza et al, 2018). The ubiquitous activity of Rabs in all aspect of intracellular membrane dynamics requires that any such pharmaceutical intervention be tightly directed to the pathologic process.…”
