Quantitative Chemical Composition of Peptidoglycan of Listeria monocytogenes

Abstract: The qualitative and quantitative chemical composition of the peptidoglycan layer of Listeria monocytogenes cell walls was determined. The peptidoglycan layer was isolated from intact cells after treatment with sodium lauryl sulfate and digestion with Pronase. The isolated peptidoglycan contained carbohydrates which included hexose, rhamnose, and amino sugars. The amino sugar was identified as glucosamine which was present in very high micromole concentration, as compared with the amino acids, muramic acid, or … Show more

“…The TEICH fractions yielded the highest values, presumably reflecting ribitol or glycerol and attached glucose or galactose residues. These carbohydrate values agree with those previously reported (10,18,19).…”

“…Ribitol was found in LCWF and decreased considerably in the preparation of PF and BHPF. Ribitol is a common constituent of teichoic acids, although teichoic acids were not detected in an analysis of serotype 4b cells (30). Ribitol has not been previously recognized in Listeria cell walls.…”

“…Glucosamine, glucose, and, most importantly, rhamnose are known to be serologically active sugars for L. monocytogenes serotype 1 (32). High quantities of rhamnose are also present in serotype 4b cells (30). No monosaccharides with molecular weights higher than those of hexoses or hexosamines were detected in any of the cell wall hydrolysates.…”

“…33 This model is useful for hydrophilic polysaccharides 69 and proteins 70 which are the major components of bacterial surfaces. 71 The contact angle (θ) is related to γ i LW , the apolar part of surface tension of condensed material (i) caused by dispersion energy between molecules, and to γ i P , the polar part of surface tension of condensed material (i) caused by dipole interaction included dipole moments and hydrogen bonds according to…”
