Quantitative Determination of Prednisone in Tablets by Infrared Attenuated Total Reflection and Raman Spectroscopy

Abstract: The quantification of prednisone in tablets was performed using partial least squares (PLS) models based on FTIR-attenuated total reflection (ATR) and FT-Raman spectra. To compare the predictive ability of these models, the relative standard error of prediction (RSEP) values were calculated. In the case of prednisone determination from the FT-Raman data, RSEP values of 3.1 and 3.2% for the calibration and validation data sets were obtained. For FTIR-ATR models, which were constructed using five spectra for eac… Show more

“…P raw material exhibited two characteristic bands in the range 1590–1710 cm −1 corresponding to symmetric and antisymmetric stretching of C = O aliphatic group [ 29 , 30 ]. The FT-IR spectrum of Zn 2+ crosslinked alginate blank beads presented bands at 1650 and 1425 cm −1 corresponding to the stretching of symmetric and antisymmetric carboxylic groups of the Zn-alginate complex and due to the coordination between the metal ion and the functional groups of the polymer [ 31 ].…”

“…Leave-one-out cross validation (LOOCV) was used to evaluate the reliability of the LDA model based on the training dataset, followed by challenging the model with the test dataset. The misclassification rates of both the training and test dataset were calculated to determine an optimal number of PCs [47]. The test dataset was rotated into a new dataset of PCs by the loadings of the PCA of the training dataset as described previously, so as to convert two datasets in the same spectral space [28].…”

“…This technique has been applied by the World Health Organization (WHO), the State Food and Drug Administration of China (SFDA) as a strategic plan for the quality assessment of TCM [9,10], and got increasing concerns because it focuses on the characterization of the total sample composition. Several ways, for instance, high-performance thin layer chromatography (HPTLC) [11][12][13][14], thin-layer chromatography (TLC) [15][16][17], infrared spectroscopy (IR) [18][19][20], mass spectrometry (MS) [21,22], gas chromatography (GC) [23,24], high-performance liquid chromatography (HPLC) [25][26][27], nuclear magnetic resonance (NMR) [28][29][30][31] and high-performance liquid chromatography (UPLC) [32][33][34] are utilized for fingerprinting to characterize multiple compounds in the tested herbal samples. Among the abovementioned methods, HPLC is generally the most applied method for its high sensitivity, selectivity, and low cost [35].…”
