Quantitative Determination of Serum Triglycerides by the Use of Enzymes

Abstract: We describe a novel method for determining serum triglycerides, in which an enzymatic hydrolysis replaces the more commonly used saponification procedure. Under the conditions of the assay, the enzymatic hydrolysis can be completed in less than 10 min by the combined action of a microbial lipase and a protease. We have been able to demonstrate complete hydrolysis of triglycerides by thin-layer chromatography of the reaction products, by recovery of glycerol from sera of known triglycerides content, and by comp… Show more

“…Total T4, T3 and TSH remained essentially unchanged post heparin administration as reported previously by us (McDougall et al, 1982). Serum NEFAs were determined by the method of Falholt et al (1973) and triglycerides by an enzyme colorimetric method (Bucolo & David, 1973) with a Technicon auto analyzer 11.…”

“…Serum cholesterol and triglycerides were assayed enzymically by a Technicon SMAC biochemical analyser (Lie ei al. 1976;Bucolo & David 1973). Lipoprotein fractions were separated electrophoretically and the relative proportions of the lipoprotein fractions assessed on agarose gel (Straw & Wurm 1958).…”

“…The total leukocytes count (WBCs count; 10 3 ml −1 ) and differential leukocyte counts were determined using the method described by Dacie and Lewis (1991). The serum samples were used to estimate the serum glucose, cholesterol and triglycerides levels colorimetrically according to the previous methods (Allain, Poon, Chan, Richmond, & Fu, 1974;Buccolo & David, 1973;Trinder, 1969) respectively.…”
