Quantitative Determination of Verbascoside inScrophularia scorodonia. by High-Performance Liquid Chromatography

Abstract: A reversed-phase high-performance liquid chromatographic method for the determination of verbascoside and an analytical study of iridoids and others phenylpropanoids in Scrophularia scorodonia L. is reported and validated. The method is applied for the analysis of stem, leaf, and flower extracts. The results showed that verbascoside represents 1.25% in leaf extracts.

“…One group of aromatic protons belongs to the caffeic acid replacement, while others belong to the phenylethanoid structure. The structure of verbascoside is well known in many reviewed literature (232425); our data are in good conformity with aforementioned studies. Existence of characteristic and typical peaks at δ 6.20 ( d , 1H, J = 15.86 Hz) and δ 7.45 ( d , 1H, J = 15.73 Hz) is the main cause for presence of phenyl propanoid structure.…”

“…Among the protons of glucose, H-4 glucose showed higher shift at 4.68 ppm, which proved the connection of caffeic acid substitution on C-4 glucose. 13 C-NMR spectrum of verbascoside was completely in agreement with published data (232425).…”

“…In general, the phytochemical compounds herein found in the two S. tenuipes fractions are in line with previous reported data for Scrophularia plants, since phenylethanoid/phenylpropanoid glycosides and iridoids represent their major metabolites. In this regard, martynoside has been previously isolated from S. dentata [1], S. xanthoglossa [24], S. koraiensis [25], and S. umbrosa [26], while the iridoid harpogoside was reported for S. ningpoensis [6], S. scorodonia [7,27], S. buergeriana [8], and S. nodosa [5]. Additionally, caffeic acid, coumaric acid derivatives, and kaempferol derivatives were also previously detected in Scrophularia plants.…”
