Quantitative Estimation of Sorafenib Tosylate Its Pure Form and in Its Tablet Formulation by RP‐HPLC Method

Abstract: A simple, accurate, specific reverse-phase, high-performance liquid chromatography method has been developed for the determination of sorafenib tosylate in its pure form and its tablets. In this method, sorafenib tosylate was eluted by isocratic mode using a Phenomenex Luna C18 column by a mobile phase composition of acetonitrile and water in the ratio of 82.5 : 17.5, v/v. The flow rate was 1.5 mL/min. The eluted drug was monitored at 265 nm and the method was found to be linear from 5 to 80 μg/mL. The method … Show more

“…Thus, factors like mobile phase ratio and flow rate were finally selected as the CMPs for subsequent optimization studies. It was observed during the screening studies that the selectivity of the method for the drug was higher at acidic pH (Venkata Rao, Ramu, Biksham, Neeharika, & Rambabu, ) and at higher column temperature (Kalaichelvi & Jayachandran, ). Thus, the pH of the mobile phase was adjusted to 4.1 using orthophosphoric acid and formic acid as the pH modifiers during HPLC and LC‐MS studies, respectively, while the column temperature was fixed at 40°C.…”

“…Several attempts have already been reported in the literature at the liquid chromatographic analysis of SFN using RP‐HPLC (Kalaichelvi & Jayachandran, ), High‐performance thin‐layer chromatography (HPTLC) (Shivaji & Pramila, ) and liquid chromatography tandem mass spectroscopy (Li et al, ), mostly employing buffers in mobile phase composition. The buffer salts, in this regard, are documented to exhibit a tendency to block the column(s) (http://www.nestgrp.com/pdf/colcare).…”
