Quantitative Real-Time PCR Detection of TRPV1–4 Gene Expression in Human Leukocytes from Healthy and Hyposensitive Subjects

Abstract: Background: Besides functioning as chemosensors for a broad range of endogenous and synthetic ligands, transient receptor potential vanilloid (TRPV) 1-4 channels have also been related to capsaicin (TRPV1), pain, and thermal stimuli perception, and itching sensation (TRPV1-4). While the expression of the TRPV1-4 genes has been adequately proved in skin, sensory fibres and keratinocytes, less is known about TRPV3 and TRPV4 expression in human blood cells.

“…TRPV1 is also expressed in the CNS, including the spinal cord, striatum, hippocampus, cerebellum, and amygdala (19)(20)(21)(22)(23). Finally, TRPV1 is found in nonneuronal cells, including leukocytes, smooth muscle cells, and endothelial cells (11,17,(24)(25)(26)(27). In this regard, TRPV1 activation has been reported to regulate the activation and proinflammatory properties of CD4 + T cells (28) NADA and TRPV1 have overlapping distributions, because NADA has been detected in the striatum, cerebellum, hippocampus, thalamus, brainstem, and dorsal root ganglia (7,19,20,23,29).…”

“…Jacob y col. 23 (2013) recomiendan el uso de más de un gen de referencia, así como también de más de un algoritmo para la evaluación de la estabilidad de la expresión. En el presente trabajo, se comparan dos metodologías (NormFinder y Rest 2009) ampliamente usadas [24][25][26][27][28][29] para evaluar siete genes de referencia candidatos, con el fin de seleccionar aquellos de mayor estabilidad para estudios de calidad seminal.…”

“…As such, it is clear that inflammatory cells have a central role in this entity. Since TRPV4 is expressed in several immune lineages (Damann et al 2009;Spinsanti et al 2008), researchers have also probed the role of TRPV4 in the cells of the innate immune system during ARDS. In a series of studies in isolated perfused mouse lungs, TRPV4 knockout resulted in decreased sensitivity to VILI (Hamanaka et al 2010).…”
