Quercetin and Coumarin Inhibit Dipeptidyl Peptidase-IV and Exhibits Antioxidant Properties: In Silico, In Vitro, Ex Vivo

Abstract: Quercetin and coumarin, two naturally occurring phytochemicals of plant origin, are known to regulate hyperglycemia and oxidative stress. The present study was designed to evaluate the inhibitory activity of quercetin and coumarin on dipeptidyl peptidase-IV (DPP-IV) and their antioxidant potential. DPP-IV inhibition assays were performed, and evaluated IC50 values of diprotin A, quercetin, coumarin, and sitagliptin were found to be 0.653, 4.02, 54.83, and 5.49 nmol/mL, respectively. Furthermore, in silico stud… Show more

“…The administration of teneligliptin, a DPP-IV inhibitor, results in a variety of beneficial effects including alleviating inflammation and oxidative stress in both the vasculature and PVAT, thus reducing atherosclerosis progression in apolipoprotein E (ApoE) knockout mice [160]. Recently, two polyphenolic compounds (coumarin and quercetin) have been suggested to inhibit DPP-IV [161]; however, further studies are needed to investigate whether polyphenolic compounds may reduce oxidative stress and target DPP-IV in PVAT.…”

“…The synthesis of different organic solvent extracts may differ in quality and concentration depending upon the difference in the polarity of the solvent used for extraction and the extracted polyphenols ( 42 , 43 ). Phenolic compounds such as gallic acid, caffeic acid, ferulic acid, trans-resveratrol, quercetin, fisetin have been shown to act as natural antioxidants by neutralizing free radicals ( 44 , 45 ).…”

“…Furthermore, DPP-IV inhibitors help enhance insulin secretion from ÎČ-cells by inhibiting the DPP-IV enzyme and increasing GLP-1 circulation in the body. Recent studies have reported the presence of protease inhibitors, such as alkaloids, flavonoids, glycosides, phenolic acids, polysaccharides, peptidoglycan, glycopeptides, and steroids, that act as DPP-IV inhibitors, along with their antioxidant properties [ 17 , 82 , 83 , 84 , 85 ]. Table 2 presents efficacious DPP-IV inhibitors with its inhibition activity (percentage/IC 50 ) from natural sources, their active parts, and medicinal uses.…”
