Rab8 and Rabin8-Mediated Tumor Formation by Hyperactivated EGFR Signaling via FGFR Signaling

Abstract: The epidermal growth factor receptor (EGFR) signaling is important for normal development, such as vulval development in Caenorhabditis elegans, and hyperactivation of the EGFR is often associated with cancer development. Our previous report demonstrated the multivulva (Muv) phenotype, a tumor model in C. elegans (jgIs25 strain) by engineering LET-23/EGFR with a TKI-resistant human EGFR T790-L858 mutant. Because Rab proteins regulate vesicle transport, which is important for receptor signaling, we screened the… Show more

“…In the exocytic transport to the PM, Rab8 and Rabin8, a GEF of Rab8, are required for fibroblast growth factor (FGF) 2 secretion to promote the EGF receptor (EGFR) signaling pathway which contributes to lung cancer malignancy [32]. In addition, Rab25 promotes or blocks tumor growth through the chloride intracellular channel protein 3 protein which is necessary for Rab25-mediated integrin recycling from late endosomes/lysosomes and drive cancer progression [33].…”

“…And there has been a promising prognosis predicting model for melanoma based on TBC family protein (Tang D. et al, 2020). Furthermore, Rab8 and Rabin8 (the GEF of Rab8) promote tumor formation (Choi et al, 2020).…”

“…As a result, osimertinib is approved for both first-line treatment of patients with T790M-positive and advanced EGFR -mutated NSCLC who have progressed on first- or second-generation EGFR-TKIs [ 15 ]. Currently, other third-generation EGFR-TKIs targeting the T790M mutation, such as Rociletinib, Olmutinib, Nazartinib, and Naquotinib are under clinical evaluation in addition to osimertinib [ 17 , 18 ].…”
