Radiation-Induced Metabolic Shifts in the Hepatic Parenchyma: Findings from 18F-FDG PET Imaging and Tissue NMR Metabolomics in a Mouse Model for Hepatocellular Carcinoma

Abstract: Purpose: By taking advantage of 18F-FDG PET imaging and tissue nuclear magnetic resonance (NMR) metabolomics, we examined the dynamic metabolic alterations induced by liver irradiation in a mouse model for hepatocellular carcinoma (HCC). Methods: After orthotopic implantation with the mouse liver cancer BNL cells in the right hepatic lobe, animals were divided into two experimental groups. The first received irradiation (RT) at 15 Gy, while the second (no-RT) did not. Intergroup comparisons over time were perf… Show more

“…The metabolomics profile in the only PMx study based on tissue samples was carried out using high-resolution magic angle spinning (HR-MAS) NMR spectroscopy [ 29 ]. Although this particular PMx study was performed using HR-MAS, a non-destructive method only requiring minimal sample preparation, some other metabolomics studies, such as those based on fecal samples, rely on a previous extraction of polar metabolites [ 35 , 36 , 37 ]. In general, the final percentage of deuterated water in samples not previously subjected to metabolite extraction (i.e., plasma, serum, saliva, urine, etc.)…”

“…The spin-echo delay is set at 300 μs and a minimum of 128 loops [ 17 ]. Our platform used a standard internal reference sodium 3-trimethylsilyl-2,2,3,3-tetradeuteropropionate (TSP) to calibrate metabolic concentrations in the aqueous phase [ 19 , 20 ]. The aqueous metabolites were identified by Chenomx NMR Suite software (Chenomx, Inc., Edmonton, AB, Canada).…”

“…A statistical analysis (one factor) module was performed with normalization by median, log transformation, and Pareto scaling in data processing for this dataset that is approximate to a normal distribution shape [ 22 ]. Metabolites satisfying three conditions: fold change over 1.2 or less than 0.8 [ 23 ], p -value in Wilcoxon rank-sum test less than 0.05 for a small number of samples, and not a full normal distribution of samples and variable importance in projection (VIP) score >1.5 in the PLS-DA model were considered significant [ 19 ].…”

“…In pulmonary arterial hypertension, metabolic reprogramming, including upregulated glutamine and altered glycine and choline metabolism, can be detected through PET imaging in mouse models (Izquierdo-Garcia et al 2018 ). Radiation-induced metabolism shifts in hepatocellular carcinoma lesions, involving increased glycolysis and impaired gluconeogenesis, were revealed using FDG-PET and metabolomics (Chung et al 2021 ). In type 2 diabetes and insulin resistance, correlations were found among PET/MRI parameters, metabolites from metabolomics profiling of subcutaneous adipose tissue and plasma, which provided new insights into the research area (Diamanti et al 2020 ).…”
