The reaction of th e primar y am ine of fumonisin B1 (FB1) with glucose was hypoth esized to detoxify th is mycotoxin. Eight y 10-day-old fema le F344/N rat s were injected intra peritoneally with dieth ylnitrosam ine (DEN; 15 mg/kg of body weig ht). At 4 wee ks of age, th e wean ed rat s were ran domly assigned to one of four tr eatm ent groups with 20 rat s each. At 9 wee ks of age, four rat s from each tr eatm ent group were kille d. At 12 wee ks, anoth er five rat s from each group were kille d. At 20 wee ks of age, th e rema ining rat s were kille d. In compar ison with th e rat s fed basal diet or FB1-glucose (conta ining 25 pp m of FB1), rat s fed 8 pp m (resid ua l amount of free FB1 in th e FB1-glucose mixtur e) or 25 pp m of FB1 ha d great er alan ine am inotran sfera se activity at 9 an d 20 wee ks of age (P < 0.001), great er endogenous hep at ic prosta glan din E2 production at 20 wee ks of age (P < 0.05), an d significant ly lower plasma cholesterol at 20 wee ks of age (P < 0.01). Placenta l glutath ione S-tran sfera se (PGST )-positive an d γ-glutam yltran sfera se (GGT)-positive altered hepatic foci (AHF) occurr ed only in rat s fed 25 pp m of FB1 at 20 wee ks of age. Hep at ic natura l kille r (NK) cell activities were similar among th e four groups, but th e percenta ge of tota l live r-associat ed mononuclear cells exhibi ting th e NKR-P1bright mark er was significant ly great er in rat s fed FB1-glucose, FB1 (8 pp m) an d FB1 (25 pp m) than in contr ol rat s at 9 wee ks of age, an d FB1-glucose-tr eat ed rat s ha d significant ly lower NKR-P1bright cells as a percenta ge of tota l live r-associat ed mononuclear cells than rat s fed 25 pp m of FB1 at 20 wee ks of age (P < 0.05). PGST-or GGT-positive AHF were not detected in an y tr eatm ent group at 9 or 12 wee ks of age. At 20 wee ks of age, ha lf of th e rat s fed 25 pp m of FB1 ha d PGSTan d GGT-positive AHF . The sp hingan ine (Sa) concentrat ion an d th e Sa/sp hingosine (So) rat io were significant ly great er in th e rat s fed 25 pp m of FB1 diet as compar ed with th e contr ol groups at , respe ctively, 12 or 20 wee ks of age. Therefore, FB -induced hep atotoxicity an d promotion of hep atocarcinogenesis . The Sa/So rat io was not th e most sensitive biomark er of FB1 toxicity. The reaction of the primary amine of fumonisin B 1 (FB 1 ) with glucose was hypothesized to detoxify this mycotoxin. Eighty 10-day-old female F344/N rats were injected intraperitoneally with diethylnitrosamine (DEN; 15 mg/kg of body weight). At 4 weeks of age, the weaned rats were randomly assigned to one of four treatment groups with 20 rats each. At 9 weeks of age, four rats from each treatment group were killed. At 12 weeks, another five rats from each group were killed. At 20 weeks of age, the remaining rats were killed. Fumonisin B1 (FB1) was reacted with fructose in an attempt to detoxify this mycotoxin. Fischer 344/N rats were initiated with diethylnitrosamine (15 mg/kg body weight) and then fed 69.3 μmol FB1/kg diet or 69.3 μmol FB1 reacted with fructose (FB1−fructose)/kg diet for 4 weeks. In comparison with the rats fed basal diet or FB1−fructose, the FB1-fed rats had significantly increased plasma cholesterol (P < 0.01), plasma alanine aminotransferase activity (P < 0.05), and endogenous hepatic prostaglandin production (P < 0.05). Placental glutathione S-transferase-positive and γ-glutamyl transferase-positive altered hepatic foci occurred only in the FB1-fed rats. Liver-associated natural killer (NK) cell activity was significantly decreased in the FB1-fed rats and increased in the group fed FB1-fructose, as compared with the basal group (P < 0.03). Therefore, modifying FB1 with fructose seems to prevent FB1-induced hepatotoxicity and promotion of hepatocarcinogenesis while stimulating liver-associated NK cell activity in rats. A B S T R A C T Cyclooxygenase inhibitors prevent the pulmonary vasomotor changes in response to low-dose endotoxin. We, therefore, explored the role of two highly vasoactive prostanoids, thromboxane A2, a vasoconstrictor, and prostacyclin, a vasodilator, in the transient pulmonary vasoconstriction and subsequent loss of alveolar hypoxis vasoconstriction (AHPV) that follows endotoxin. AHPV was tested in the dog with a double-lumened endotracheal tube allowing ventilation of one lung with nitrogen as a hypoxic challenge while the other lung was ventilated with oxygen to maintain systemic oxygenation. Relative distribution of perfusion to the two lungs was assessed with intravenous 133Xe and external scintillation detectors. The stable metabolites of thromboxane and prostacyclin, i.e., thromboxane B2 and 6-keto-prostaglandin F1,a were measured in plasma with radioimmunoassay. 15 ,ug/kg i.v. ofendotoxin induced no rise in pulmonary vascular resistance (PVR), but prevented AHPV so that the initial 33% (±2 SEM) decrease in perfusion to the hypoxic lung became only a 2% (± 1) decrease. Circulating levels of thromboxane and prostacyclin concurrently rose (P < 0.01) from nondetectable levels to 380 pg/ml (±40) and 360 pg/ml (±130). 150 ,g/kg of endotoxin induced a transient rise in PVR from 4.09 to 9.00 mm Hg/liter per min in association (r = 0.89, P < 0.01) with a sharp rise in thromboxane levels to 4,460 pg/ml (± 1,350) whereas prostacyclin levels were elevated less markedly to 550 pg/ml (+400). Prostaglandin F2<,X another vasoconstrictor, was not elevated. 30 min after endotoxin when PVR was again base line and AHPV lost, thromboxane fell significantly (P < 0.01) to 2,200 pg/ml (± 1,100) whereas prostacyclin remained elevated at 360 pg/ml (±135), a level similar to that seen when 15 ,g/kg of endotoxin induced Dr. Peterson is an Established Investigator ofthe American Heart Association.
