Radioimmunoassay of Serum Deoxycorticosterone: Normal Values and Changes of Serum Deoxy-corticosterone after Adrenal Stimulation

Abstract: A sensitive radioimmunoassay for estimating serum deoxycorticosterone is described which involves descending "overrunning" paper chromatography. An ethanolic paper eluate is used as a solvent for unlabelled deoxycorticosterone in the preparation of the standard curve. The values of the water blanks (7 ng/1) are similar to the sensitivity of the standard curve (5.8 ng/1). The normal mean serum concentration of deoxycorticosterone was found to be 84 ± 29 ng/1 (n = 30) in males and 55 ± 31 ng/1 (n = 35) in female… Show more

“…The association constant for the DOC-antibody complex was calculated to be 1.5 x 10 5 M~'s~' and the dissociation constant to be 5 x 10~5s~' (5). The coefficients of variation for replicate interassay determination at low, medium and high DOC-concentrations amounted to 19.5 and 21.8% and for intraassay determination to 8.6 to 17.8%.…”

“…The results show that, even though hormone production by metastases may influence TSH secretion to some extent, TSH levels after withdrawal of the replacement therapy are mainly related to the individual pituitary response rather than to the presence or absence of metastases. However, an increase of TSH levels after 131 ropin (TSH); they are able to concentrate radioiodine and often to synthesize thyroxine (T 4 ) and triiodothyronine (T 3 ) (1-3); in rare cases thyroid hormone production by thyroid carcinomas may cause overt hyperthyroidism (4)(5)(6).…”

“…3 mg corticosterone/day in young men (Romanoff & Baxter 1975) or 100-200 mg/day for aldosterone (Katzung 2004, Goodman et al 2007); other authors give higher values for DOC, e.g. (Harris et al 1967), and its plasma concentration is typically 200-400 pmol/l (Brown & Strott 1971, Oddie et al 1972, Schoneshofer et al 1975. Its secretion may be enhanced by ACTH and by angiotensin II and potassium ions, suggesting both a zona fasciculata and a zona glomerulosa origin (Brown et al 1972b), and it is also inevitably enhanced in conditions in which 11b-hydroxylase (CYP11B1) has reduced activity.…”

“…12 Deoxycorticosterone is not secreted from the adrenal glands in large amounts so plasma concentrations are low compared to other adrenal hormones, approximately 150-300 pmol/L. [13][14][15] Due to this very low concentration in healthy people, there are not likely to be many occasions when this steroid is detected. The bloodspot 17OHP assay is designed for monitoring CAH patients, 90-95% of whom will have 21-hydroxylase deficiency.…”
