Rapid assignment of the swine major histocompatibility complex (SLA) class I and II genotypes in Clawn miniature swine using PCR‐SSP and PCR‐RFLP methods

Abstract: The combination of PCR-SSP and PCR-RFLP methods facilitate the rapid identification of the three haplotypes and SLA class I and II homozygotes in individual Clawn miniature swine.

“…In addition, swine are the only large animal that have inbred MHC-defined strains available [19], [20], [21]. One such strain, Clawn miniature swine, was established in 1978 by Nakanishi and colleagues at Kagoshima University, and its swine leukocyte antigen (SLA) genotype was identified [20], [22], [23]. Previously, we established iPS cells from inbred SLA-defined Clawn miniature swine [24] (Fig.…”

“…Dev. 58: [398][399][400][401][402][403] 2012) C lawn miniature pigs (inbred strain) have a potential value for biomedical research and xenotransplantation due to their clear genetic background [1][2][3] and few individual differences; thus, they have stimulated investigation of novel strategies directed at generating a genetically modified miniature pig by somatic cell nuclear transfer (SCNT). We have shown how to produce live offspring derived from Clawn miniature pig SCNT embryos; however, the efficiency still remains low [4].…”

“…In addition, the haplotypes of swine leukocyte antigens, which are important immunogens for humoral responses and important mediators of the cellular immune responses, are well defined in these miniature pigs [32]. This means that α-1,3-GT knockout Clawn miniature pigs are useful animals as a source of organs for xenotransplantation.…”
