Nanoparticles have attracted much attention as a key material for new biomedical and pharmaceutical applications. For success in these applications, the nanoparticles are required to translocate across the cell membrane and to reach to inside of the cell. Among several translocation pathways of nanoparticles, the direct permeation pathway has a great advantage due to its high delivery efficacy. However, despite many research efforts, key properties and factors for driving the direct permeation of nanoparticle and its underlying mechanisms are far from being understood. In this article, experimental and computational studies regarding the direct permeation of nanoparticles across a cell membrane will be reviewed. Firstly, experimental studies on the nanoparticle-cell interactions, where spontaneous direct permeation of nanoparticles was observed, are reviewed. From the experimental studies, potential key physico-chemical properties of nanoparticles for their direct permeation are discussed. Secondly, physical methods such as electroporation and sonoporation for delivering nanoparticles into cells are reviewed. Current status of technologies for facilitating the direct permeation of nanoparticle is presented. Finally, we review molecular dynamics simulation studies and present the latest findings on the underlying molecular mechanisms of the direct permeation of nanoparticle.

Raman spectroscopy is an increasingly popular technique in many areas including biology and medicine.It is based on Raman scattering, a phenomenon in which incident photons lose or gain energy via interactions with vibrating molecules in a sample. These energy shifts can be used to obtain information regarding molecular composition of the sample with very high accuracy. Applications of Raman spectroscopy in the life sciences have included quantification of biomolecules, hyperspectral molecular imaging of cells and tissue, medical diagnosis, and others. This review briefly presents the physical origin of Raman scattering explaining the key classical and quantum mechanical concepts. Variations of the Raman effect will also be considered, including resonance, coherent, and enhanced Raman scattering. We discuss the molecular origins of prominent bands often found in the Raman spectra of biological samples. Finally, we examine several variations of Raman spectroscopy techniques in practice, looking at their applications, strengths, and challenges. This review is intended to be a starting resource for scientists new to Raman spectroscopy, providing theoretical background and practical examples as the foundation for further study and exploration.

Understanding uptake of nanomaterials by cells and their use for intracellular sensing is important for studying their interaction and toxicology as well as for obtaining new biological insight. Here, we investigate cellular uptake and intracellular dynamics of gold nanoparticles and demonstrate their use in reporting chemical information from the endocytotic pathway and cytoplasm. The intracellular gold nanoparticles serve as probes for surface-enhanced Raman spectroscopy (SERS) allowing for biochemical characterisation of their local environment. In particular, in this work we compare intracellular SERS using non-functionalised and functionalised nanoparticles in their ability to segregate different but closely related cell phenotypes. The results indicate that functionalised gold nanoparticles are more efficient in distinguishing between different types of cells. Our studies 2 pave the way for understanding the uptake of gold nanoparticles and their utilisation for SERS to give rise to a greater biochemical understanding in cell-based therapies.
