Rapid detection and quantitation of ketamine and norketamine in urine and oral fluid by wooden-tip electrospray ionization mass spectrometry

Abstract: Drug analysis is an indispensable task in controlling drug abuse, which is a serious problem worldwide nowadays. In this study, we report a simple and rapid approach for detection and quantitation of drugs-of-abuse in urine and oral fluid by wooden-tip electrospray ionization mass spectrometry (WT-ESI-MS). We demonstrated that ketamine, one of the most common abused drugs, and its major metabolite, norketamine, in raw urine and oral fluid could be readily detected and quantified by WT-ESI-MS with only little s… Show more

“…Samples were loaded by pipetting onto the tip or dipping the tip into the sample solutions. This technique have been used for analysis of pharmaceuticals [58], ketamine and norketamine in urine and oral fluid [59], and Chinese herbal medicine preparations [60][61][62]. It has also been shown to act as a chromatographic column for separation of sample components, such as salts and proteins [63].…”

“…Wooden toothpicks have been frequently used as the substrates due to low cost and disposable features. 64) is approach has been applied for various applications, such as analysis of toxic substance in food samples, 65) drugs in urine and oral uid, 66) and metabolite ngerprinting. 67) In addition, wooden tip-based ESI was demonstrated for rapid analysis of pharmaceutical samples in various forms, including tablets, granules, capsules, suspensions and oral liquids.…”

“…Sequential and exhaustive ionization was also observed in wooden-tip ESI with an order di erent from that in PESI, 22) enabling separated ionization of salts and proteins thus direct analysis of protein samples with high salt content by the technique. 10) More recently, we demonstrated the application of woodentip ESI in direct detection and quantitation of drugs-ofabuse in raw oral uid and urine with acceptable sensitivity, linear range, accuracy and precision 42) (Figs. 4(c) & (d)).…”
