Rapid detection of natriuretic peptides by a microfluidic LabChip analyzer with DNA aptamers: Application of natriuretic peptide detection

Abstract: Rapid detection of brain natriuretic peptide (BNP) concentration can be used for the diagnosis of acute heart failure and for the evaluation of the effectiveness of a clinical therapy. We used the systematic evolution of ligands by exponential enrichment method to develop DNA aptamers for BNP whose sequences were determined by cloning method and consensus sequence analysis. A total of eight conserved sequences was identified. By combining the fluorescent-labeled aptamers with fast protein lab-on-chip analysis,… Show more

“…The value for the interaction between BNP and antiBNP agrees well with previous reports. 25 The DNA aptamer sequence was recently identified by Lin et al, 26 and this is the first time a binding affinity coefficient has been reported. A detection limit of ∼1 nM shown here is typical for conventional SPR measurements.…”

“…Simultaneous determination of angiotensin II and angiotensin‐(1–7) in plasma and urine from normotensive and hypertensive rats, with and without antihypertensive treatment, was possible over the range 0.01–500 pmol/mL within 10 min by CZE with UV‐detection at 200 nm in the BGE composed of 10 mM boric acid, 3 mM tartaric acid, 10 fM AuCl 3 , pH 9.80, at +30 kV in 75 μm id×57/50 cm long FS capillary 216. Rapid determination of brain natriuretic peptide in blood, biomarker for the diagnosis of acute heart failure and for the evaluation of the effectiveness of a clinical therapy, was possible by microfluidic LabChip analyzer using fluorescently labeled aptamers 217.…”

“…Another excellent work using a highly specific and sensitive DNA aptamer, instead of using antibodies, for the brain natriuretic peptide (BNP) assay has been reported by using Agilent lab-on-a-chip (Agilent Technologies, Waldbronn, Germany) technology [ 52 ]. BNP is a neurohormone that causes diuresis, natriuresis and vasodilatation.…”
