Rapid Detection of Rongalite via a Sandwich Lateral Flow Strip Assay Using a Pair of Aptamers

Abstract: A sandwich lateral flow strip assay (LFSA) using a couple of aptamers functionalized with gold nanoparticles (AuNPs) was designed to assess the presence of rongalite in agrifood products. More specifically, a biotin-labeled primary A09 aptamer immobilized on a streptavidin-coated membrane and a secondary B09 aptamer conjugated with AuNPs were developed as capturing and signaling probes, respectively. This system allows the successful and direct detection of rongalite in food samples with concentrations as low … Show more

“…To achieve a selective detection, one simply needs to optimize an appropriate signaling antibody that will pair with GAGs. The need for one antibody instead of two (as required for constructing traditional sandwich-type LFSA sensors) will significantly shorten the screening time 47,48 when applied toward a new pathogen or variant, as well as drastically lowering the cost, potentially 10-fold, as compared to current LFSA technologies. 98,99 GlycoGrip has remarkable promise as a widespread tool for capturing and detecting current and emerging viruses.…”

“…Furthermore, in the case of LFSA sandwich-type detection, pairs of antibodies or aptamers must be screened for both capturing and reporting, which delays sensor design and development. 47 , 48 In contrast, GAGs could serve as universal capture agents for various viral analytes including mutant strains, resulting in an easily generalizable LF platform. This could greatly reduce the assay’s antibody screening and optimization time, which typically takes anywhere from 12 to 16 weeks.…”

“…Not only this, rongalite is an attractive building block to assemble sultine derivatives which are useful masked dienes partners for Diels‐Alder (DA) reaction to construct intricate molecules which are otherwise challenging or almost impossible by other means . On the other hand, very recently Han and co‐workers have revealed a promising role of aptamers for the detection of rongalite in food with high sensitivity and specificity through sandwich‐type lateral flow strip and enzyme‐linked aptamer assays . Therefore, keeping the above stated potential applications of this valuable reagent in mind, herein I have tried to disclose its applicability not only in modern synthetic organic chemistry but also its utility in material sciences.…”

“…Recently, several direct methods that adopted rongalite itself as the detection maker were reported as alternative methods for the rapid detection of rongalite. For instance, based on a rongalite-specific aptamer, Li et al established a sandwich lateral flow strip (LFIA) assay with the limit of detection (LOD) of 1.0 μg/mL [ 15 ]; Jing et al developed a sandwich-type enzyme-linked immunosorbent assay (ELISA) with the LOD of 0.57 ng/mL ( Table S1 ) [ 16 ]. Although the aptamer-participated methods were rapid and sensitive, the key reagent aptamer with high specificity and affinity was hard to be prepared.…”
