Rapid determination of rosmarinic acid and its two bioactive metabolites in the plasma of rats by LC–MS/MS and application to a pharmacokinetics study

Abstract: Rosmarinic acid (RA), an ester compound of caffeic acid (CA) and 3,4‐dihydroxyphenyllacic acid, is widely distributed in the herbs of the Lamiaceae family and has shown a wide spectrum of pharmacological properties. CA and FA (ferulic acid) are two bioactive metabolites in vivo after oral administration of RA; however, a rapid and robust analytical approach that can enable the quantitative assay of RA and two bioactive metabolites is still lacking. A liquid chromatography/tandem mass spectrometry method was es… Show more

“…Subsequently, the absorption of RA gradually increased, reaching its peak concentration after 2 hours. These findings align with the observation reported by Wang et al , 25 who observed rapid absorption and intermediate-speed elimination of RA in rats following oral administration. The gradual decline in plasma levels of RA suggests extensive biotransformation through phase I and phase II metabolism in the liver.…”

“…They efficiently facilitated the transfer of acyl groups from acyl donors to shikimate or quinate, resulting in three p -coumaroyl shikimate isomers, three caffeoyl shikimate isomers, and three p -coumaroyl quinate isomers in our tests ( Figure 2 A,C,D). Moreover, they also catalysed the formation of p -coumaroyl-4’-hydroxyphenyllactate (a precursor of RA) by accepting the substrate pair consisting of p -coumaroyl-CoA and 4-hydroxyphenyllactate, as confirmed through precise molecular ion mass and tandem mass spectrometry (MS 2 ) analysis ( Figure 2 B), in accordance with previous studies [ 31 , 32 , 33 ]. In the case of Ml AT2, only trace products were detected via mass spectrometry spectrum when using p -coumaroyl-CoA as the acyl donor.…”

“…Compound (6) was detected at Rt 4.96 min with m/z 359.0782 in all of the studied taxa except for O. latifolia. It gave fragmentation pattern of m/z 197, 179, 161 that was in compliance with that of rosmarinic acid (Wang et al, 2021).…”
