Rapid diagnosis of micrometastasis of gastric cancer using reverse transcription loop-mediated isothermal amplification

Abstract: Abstract. Methods to detect metastases in biopsy specimens with certain rapidity and accuracy are essential to performing tailor-made surgeries for solid malignancies. Reverse transcription loop-mediated isothermal amplification (RT-LAMP) reaction is a novel technique for detecting mRNA expression of target sequences with high sensitivity and rapidity, even from crude samples without RNA purification. Applicability to detect lymph node (LN) micrometastasis of gastric cancer was tested. Total of 26 LNs were ret… Show more

“…There are quite many studies where patients that were node negative both with HE and anti-CK immunoassay, cancerous load was finally visible when rt-PCR was used ( 11 ). The sensitivity of rt-PCR is outstanding (>90%) ( 12 ), but is also demanding in time so it cannot be used in everyday clinical practice, plus it has false positive results. Undoubtedly, it is a promising method for the future, where minimal invasive surgery combined with individualized lymphadenectomy will probably prevail.…”

“…A significant portion of SLM (reports range 10 to 60%) ( 3 , 5 , 6 , 9 , 12 , 21 ) is not detected on level 1 nodes, but in level 2 nodes leading to so-called skip metastasis. SM usually occur along the left gastric artery, anterior common hepatic artery but can also be cited along coeliac or splenic artery.…”

“…The greatest advantage of RT-PCR is its sensitivity, such that 1 tumour cell in 1×10 7 cells can be detected [ 19 ]. The introduction of RT-PCR increased the rate of detection of LNM to a level higher than that achieved using IHC [ 20 ].…”
