Rapid Diagnosis of TGEV‐Like Coronavirus in Fattened Pigs by Indirect Immunofluorescence Labelling in Nasal Cells

Abstract: Diagnosis of TGEV-like coronavirus was made by an immunofluorescence technique using monoclonal antibodies directed against TGEV. Six, 15 weeks-old pigs were inoculated intratracheally with the strain P6008ST4. No clinical sign was observed during the course of the infection. Viral antigens were detected in the cytoplasm of nasal cells obtained from swab in five animals as early as one day p. i. and during several days. The sensitivity and specificity of this method appeared to be comparable to virus isolation… Show more

“…The strain isolated in France (2) was used. In a previous experiment (7), nasal swabs were taken from infected SPF pigs. “…9 Older weaned pigs, however, showed no impaired weight gain after PRCV exposure. 2 Despite the transient reduced weight gain, the PRCV-exposed piglets recovered from the infection over the 12-month course of this experiment and suffered no long-term ill effects from their exposure to PRCV as neonates.…”

“…PRCV was isolated in cell culture for up to 10 days after inoculation but not by 14 days after inoculation of experimentally infected pigs. 2,10 Pig-to-pig transfer of the virus, however, is usually a more sensitive method used to demonstrate virus shedding, but no such studies have been carried out for PRCV-infected pigs. In this experiment, an upper limit for PRCV secretion was established by commingling age-matched sentinel pigs with the PRCV-primed pigs.…”

“…PRCV principally multiplies in the respiratory tract, infecting epithelial cells and alveolar macrophages (Pensaert and Cox, 1989). Thus, PRCV has been detected in nasal swabs of experimentally infected pigs (Onno et al, 1989;Wesley et al, 1990) but not in the faeces, as is typical for TGEV infections. Microscopic examination of lungs reveals that PRCV causes a diffuse interstitial pneumonia (O'Toole et al, 1989;van Nieuwstadt and Pol, 1989;Cox et al, 1990).…”
