Rapid Identification and Differentiation ofBartonellaSpecies Using a Single-Step PCR Assay

Abstract: Five species of Bartonella have been reported to infect humans and cause a variety of diseases that can be difficult to diagnose. Four species of Bartonella have been reported to infect cats and dogs, and two of these species are considered to be zoonotic pathogens. Diagnosis of Bartonella infections is hampered by the slow, fastidious growth characteristics ofBartonella species. We report on the development of a single-step PCR-based assay for the detection and differentiation of medically relevant Bartonella… Show more

“…Samples were screened to determine exposure to pathogens using assays performed and interpreted following standardized protocols, as previously described , 2002, Jensen et al 2000, Troyer et al 2005, Franklin et al 2007b, Veir et al 2008, Ruch-Gallie et al 2011. In the majority of cases, pathogen exposure was determined by serological analysis.…”

“…The primers BhCS.781p and BhCS.1137n were used for amplifying the partial fragment (approximately 390 bp) of the gltA gene of Bartonella species (Norman et al, 1995). The PCR-positive samples for the gltA gene were further analyzed by PCR of the 16S/23S rRNA intergenic spacer region (ITS) (Jensen et al, 2000). For further phylogenetic analysis, confirmed Bartonella isolates were analyzed by partial sequences of the ftsZ, rpoB and ribC genes.…”
