Rapid immunohistochemical detection of tumor cells in gastric carcinoma

Abstract: Abstract. The detection of single tumor cells or tumor cell clusters represents an important issue in intraoperative frozen section analysis. For example, surgical margins may be evaluated in order to minimize the number of additional operations. Furthermore, intraoperative diagnosis of lymph node micrometastasis (LNM) may help to define the area of appropriate lymph node dissection. In addition to haematoxylin and eosin (H&E)-stained sections, immunohistochemical detection of single tumor cells or cell cluste… Show more

“…However, its application to effusion cytology is not common, especially in fluorescence ICC [18,19,20,21]. In this study, we conducted the rapid procedure for fluorescence ICC using Ber-EP4 that can be performed within 20 min using the liquid-phase method.…”

“…For example, the new EnVision TM system has been described as a very sensitive and rapid detection system for IHC [ 24 , 27 ], and Kämmerer et al and Mönig et al reduced the time for immunostaining of frozen sections to less than 13 min using the EnVision system. To do so, however, they applied a higher concentration of primary antibody than is used for standard IHC [ 13 , 17 ]. By contrast, our ultrarapid AC electric field IHC procedure required about the same amount of time as the EnVision TM system, and we were able to use the same concentration of primary antibody used in the standard IHC procedure.…”

“…Finally, we considered whether this method of immunofluorescence staining with mixing leads to reduce costs relative to conventional staining methods. To date, several methods for effective immunostaining have been used, including microwave 15 16 , ultrasound 17 , and high quality reagents 21 . Past reports focused on the reduction in staining time, and did not devote attention to reduction of cost.…”
