Rapid MALDI-TOF-MS Detection of Anthocyanins in Wine and Grape Using Different Matrices

Abstract: Matrix-assisted laser desorption/ionization timeof-flight mass spectrometry (MALDI-TOF-MS) as a new valuable technique has been used for qualitative screening of anthocyanins in wine and grape samples from Vranec and Merlot variety. For that purpose, different MALDI matrices have been tested, such as α-cyano-4-hydroxycinnamic acid, sinapic acid, 2,5-dihydroxybenzoic acid (2,5-DHB), and C 70 fullerene. It was observed that 2,5-DHB was superior with respect to all the matrices used in the study. Furthermore, ful… Show more

“…Various ionisation methods such as electrospray ionisation (ESI) [74,[89][90][91][92], atmospheric pressure chemical ionisation (APCI) [77], continuous flow fast atom bombardment (CF-FAB) [79] and matrix assisted laser desorption ionisation (MALDI) [72] have been used. Quadrupole mass analysers are most widely applied, due to their robustness and low cost.…”

“…Especially in the last 2 decades, mass spectrometry (MS) has also found extensive application in anthocyanin analysis, with ionisation methods such as matrix assisted laser desorption ionisation (MALDI) [72,73], electrospray ionisation (ESI) [74,75], atmospheric pressure chemical ionisation (APCI) [76,77] [78,79] having been applied. The main focus of the next section will therefore be on LC and LC-MS analysis of anthocyanins.…”

“…Wine characterization of Macedonian wines has been performed, including determination of phenolic and aroma compounds in commercial wines, as well as wines produced by different winemaking procedures (Ivanova et al (2009(Ivanova et al ( , 2011a(Ivanova et al ( , 2011b(Ivanova et al ( , 2011c(Ivanova et al ( , 2012). But, til now, low number of papers is dealing with determination of heavy metals in wines (Ivanova-Petropulos et al 2013), especially for Cd and Pb which have been separately determined in Macedonian wines and published in separate papers in 2006 and 2007 (Cvetković et al 2006;Karadjova et al 2007).…”

“…In addition, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) is a powerful new technique that has a great potential in food analysis, as well as on wine and grape analysis (Sugui et al 1998(Sugui et al , 1999Reed et al 2005;Carpentieri et al 2007;Ivanova et al 2011b). This technique allows successful determination of the molecular weights in complex samples directly from ion abundances in the mass spectrum without previous isolation or cleanup of the sample (Hanton 2001;Ivanova et al 2011b).…”

“…This technique allows successful determination of the molecular weights in complex samples directly from ion abundances in the mass spectrum without previous isolation or cleanup of the sample (Hanton 2001;Ivanova et al 2011b). The MALDI technique, for the first time, was demonstrated by Karas et al 1987, originally being developed for the analysis of large biomolecules, such as proteins.…”
