Rapid Screening of Taxol Metabolites in Human Microsomes by Liquid Chromatography/ Electrospray Ionization-Mass Spectrometry

Abstract: Liquid chromatography with an electrospray ionization (ESI) interface has been applied to study the anticancer drug taxol and its metabolites after incubation with human hepatic microsomes. The parent drug and its metabolites were monitored in the positive-ionization mode. Since ESI gave only quasi-molecular ions for taxol and its analogues, collision-induced dissociation experiments were carried out in order to generate fragment ions, by increasing the cone voltage at the ESI source. The product-ion mass spec… Show more

“…During the past decade, liquid chromatography/mass spectrometry (LC/MS) has been recognized as the most appropriate analytical technique for structural assignment of taxanes in complex samples due to its specificity and structure‐characterization ability 16–24. However, rapid metabolic profiling of yew materials is still a challenge, since there are numerous trace constituents from different classes and many analogues with similar chromatographic behavior, as well as many regio‐ and stereoisomers 25–29.…”

“…During the past decade, liquid chromatography/mass spectrometry (LC/MS) has been recognized as the most appropriate analytical technique for qualitative analysis or pharmacokinetic studies of taxane analogues from realistic samples due to its high sensitivity, specificity and structure characterization ability 12–24. Among these reports, electrospray ionization (ESI) was preferred due to its high‐efficiency ionization 18–24. A particular challenge for mass spectrometry is distinguishing and characterizing isomers.…”

“…The combination of LC with matrix-assisted laser desorption ionization (MALDI) started to emerge in the mid-1990s 18,19 and continuous effort was undertaken to further improve the technique [20][21][22][23][24][25] . Very recently LC-MS methods were described using desorption electro spray ionization (DESI) 26 and direct analysis in real time (DART) 27,28 interfaces.…”
