RCC1 regulates inner centromeric composition in a Ran-independent fashion

Abstract: RCC1 associates to chromatin dynamically within mitosis and catalyzes Ran-GTP production. Exogenous RCC1 disrupts kinetochore structure in Xenopus egg extracts (XEEs), but the molecular basis of this disruption remains unknown. We have investigated this question, utilizing replicated chromosomes that possess paired sister kinetochores. We find that exogenous RCC1 evicts a specific subset of inner KT proteins including Shugoshin-1 (Sgo1) and the chromosome passenger complex (CPC). We generated RCC1 mutants that… Show more

“…Pim1 is the yeast homolog of the metazoan RCC1. Given that RCC1 is implicated in regulating centromeric composition in vitro (Zhang et al, 2018), we examined the association of Pim1 with the centromeric regions of wild‐type and nup132 Δ meiotic cells. Using ChIP‐qPCR, Pim1 was detected at centromeric loci, and its localization at centromeres was significantly reduced in the absence of Nup132 (Figure 7a).…”

“…Bub1 is precociously loaded onto the meiotic centromeres in the nup132 Δ meiotic cells (Yang et al, 2015), suggesting that Nup132 regulates kinetochore dynamics via Top2 SUMOylation. In addition, studies using Xenopus egg extract indicate that increasing the level of chromosome‐associated RCC1 reduces the degree of Bub1 occupancy at the centromeres (Arnaoutov & Dasso, 2003; Zhang et al, 2018). It is possible that the reduced chromosomal binding of Pim1 allows for the untimely localization of Bub1 at the centromeres in the nup132 Δ mutant.…”

“…While these pathways seem to be distinct [Sampath et al., 2004; Maresca et al., 2009], relationships exist between the two. For instance, RCC1 acts at the kinetochore to modulate the spindle assembly checkpoint [Arnaoutov and Dasso, 2003], where it regulates the metaphase‐anaphase transition through the displacement of Sgo1 and CPC [Zhang et al., 2018]. In addition, the stabilisation of k‐fibre minus‐ends is ensured by a Ran‐GTP‐dependent mechanism [Meunier and Vernos, 2011] and Ran‐GTP is crucial for the maintenance of stable kinetochore‐MT attachments through the regulation of Aurora B [Lee et al., 2012].…”
