Reaction of sugarcane varieties to orange rust (Puccinia kuehnii) and methods for rapid identification of resistant genotypes

Abstract: Sugarcane orange rust was recently introduced into Brazil and its control is based on the use of resistant varieties. This study aimed to determine the reaction of Brazilian sugarcane varieties to the disease in the field and to compare artificial inoculation methods. Rust severity was assessed in 17 varieties at a 15-day interval. The maximum disease severity (MS%) and the area under the disease progress curve (AUDPC) were determined for each genotype. The artificial inoculation methods tested were: spraying … Show more

“…A estratégia mais empregada de controle da ferrugem alaranjada em todo mundo é o uso de variedades resistentes (10,17). Além da correta avaliação da reação do hospedeiro, a exata noção da diversidade do patógeno também é fundamental (12).…”

“…Approximately 30 mL of the spore suspension was sprayed onto the abaxial portion of the leaves of each plant using a manual sprayer for regular distribution of small droplets. In the first 24 h after inoculation, the plants were maintained in a chamber equipped with air humidifiers that were switched on for 15 min every 2 h to ensure optimal conditions of temperature and humidity (Chapola et al, 2016).…”

“…The presence of sporulation was confirmed using a 910 hand lens and the development of symptoms was monitored weekly from the time at which the first symptoms appeared until the termination of the experiment. The visual evaluation of the inoculated plants was based on a foliar chart scale for estimating the percentage of leaf area with symptoms (Ara ujo et al, 2013;Chapola et al, 2016). At the end of the experiment, disease severity of the whole canopy (percentage of leaf area with symptoms per plant) was also estimated by calculating the ratio of the leaf area with symptoms to the symptomless leaf area.…”

“…With regard to orange rust, 68 cultivars have shown distinct responses when infected with P. kuehnii strains [49]. 69 Primarily, studies in orange rust focused on molecular detection of P. kuehnii by using 70 primers that amplify ITS regions [17], the characterization of pathogen developmental 71 structures during infection [50] or the assessment of infection degree in sugarcane 72 cultivars [18,19]. Yang and collaborators [46] detected three QTL associated with 73 resistance against orange rust.…”

“…Our 82 work thus aims to study a time series of transcriptional changes in susceptible sugarcane 83 infected by P. kuehnii. 84 Materials and methods 85 Plant Material 86Plants of the sugarcane cultivar SP89-1115, susceptible to orange rust [19,49,52], were 87 grown in 500 mL plastic cups containing sterilized substrate in a greenhouse during 40 88 days and fertilized with an ammonium sulfate solution (30 g/L) ten days before 89 inoculation. One day before inoculation, spores of P. kuehnii were obtained using a 90 vacuum pump (Prismatec 101) from the abaxial side of infected leaves from Centro de 91 Tecnologia Canavieira (Piracicaba, São Paulo, Brazil) that contained open pustules.…”
