Reactive Astrocytes in Glioblastoma Multiforme

Abstract: Despite the multidisciplinary integration in the therapeutic management of glioblastoma multiforme (GBM), the prognosis of GBM patients is poor. There is growing recognition that the cells in the tumor microenvironment play a vital role in regulating the progression of glioma. Astrocytes are an important component of the blood-brain barrier (BBB) as well as the tripartite synapse neural network to promote bidirectional communication with neurons under physiological conditions. Emerging evidence shows that tumo… Show more

“…In the same picture, ion channels and ion transporters, expressed at the membrane of both astrocytes and GBM cells might then form a bridge between the two cellular types, thus enhancing tumor progression and therapeutic resistance, as suggested by a recent review (Guan, Hasan, Maniar, Jia, & Sun, ). Also, the GBM‐derived extracellular vesicles (EVs) might significantly alter the intracellular signaling and cytokine profile of normal human astrocytes to push them towards a tumor supporting behavior (Oushy et al, ).…”

“…The different expression of ARG1 and CDH1 in our model might display the limitations of our model but might also indicate that these characteristics are dependent on other components of the TME. [12,22] By implementing other cell types in our model, for instance glioma-associated astrocytes, which so far have not been fully investigated for their contribution to GBM, [23] and by gradually increasing the complexity of this model, it might be possible to clearly and specifically define the function and contribution from each TME component. To further confirm the clinical significance of the aforementioned genes, we analyzed the survival of 577 GBM patients.…”

“…Differential expression of hEAG1 and hERG1 is found amongst gliomas conferring to the malignant status and nature of the tumour. Kv11.1 is associated with abnormal expression in hGG [ 85 ]. Similarly, when supressed by siRNA hERG mediated apoptosis in glioma cell lines demonstrating a key role in glioma apoptosis [ 86 ].…”
