Real time and online dynamic speckle assessment of growing bacteria using the method of motion history image

Abstract: This paper reports the application of the motion history image (MHI) method for biospeckle processing of a bacterial growth. The method avoids the complexity as well as the large computation in sequence-matching-based methods and detects whether the speckle structure has changed or not. Encouraging experimental results on the real-time evolution of the growing bacteria during 12 h demonstrate the effectiveness of the proposed method. The MHI presented an online result without loss of resolution and definition.… Show more

“…8c ). This framework, while aimed at FLN, could easily be applied to other soil pests such as microbes or fungi, which have already been shown to be detectable by BSL 30 , 32 , 126 . Furthermore, the biocompatibility of Ludox® and easy manipulation of chemical properties in terms of nutrients and pH of transparent soil make it a suitable substrate for a wide range of soil-dwelling microorganisms.…”

“…Recently, light sheet based techniques such as SPIM (Selective Plane Illumination Microscopy) have shown great potential for live imaging studies 24 28 , but light scattering itself may not allow easy separation of nematode and root in the image 29 . On the other hand, techniques based on the dynamic light scattering created by living organisms, termed biospeckle, have shown potential as a simple, rapid and non-invasive means of tracking biological activity 30 32 . Biospeckle has the advantage of enhancing the contrast between living organisms from their abiotic background.…”

“…Biological samples show this behavior when they are illuminated with a laser. However, there are other dynamic processes where this behavior is also observed [46]. As stated, during phase separation, the domain surfaces are changing due to coalescence, thus giving dynamic surfaces.…”

“…It is used to characterize movements within a row of images in time. The MHI indicates a static pattern that the pixel intensity is a function of the recent image sequence motion and contains the fundamental information in order to determine the motion of the object during the activity 45,46 . MHI has been used profoundly for visualizing the perfusion changes locations during different stimuli 47 .…”
