Receptor-Mediated and Fluid-Phase Transcytosis of Horseradish Peroxidase across Rat Hepatocytes

Abstract: Horseradish peroxidase (HRP) is often used as a fluid-phase marker to characterize endocytic and transcytotic processes. Likewise, it has been applied to investigate the mechanisms of biliary secretion of fluid in rat liver hepatocytes. However, HRP contains mannose residues and thus binds to mannose receptors (MRs) on liver cells, including hepatocytes. To study the role of MR-mediated endocytosis of HRP transport in hepatocytes, we determined the influence of the oligosaccharid mannan on HRP biliary secreti… Show more

“…Dextran labeled with FITC or other fluorophores is generally applied as a fluid phase marker, especially to determine the involvement of macropinocytosis in the internalization of cargo . Although we do not consider that dextran in a complex with protamine, DNA and SLN functions as a fluid phase marker, recent published data demonstrate that the dextran alters the properties of protamine‐DNA‐SLN complexes with respect to erythrocyte agglutination .…”

“…Mannose receptors have been reported to be constitutively expressed by liver macrophages, endothelial cells and hepatocytes (Kempka and Kolb-Bachofen, 1988; Ellinger and Fuchs, 2010). However, while mannose receptor expression is upregulated in liver macrophages and endothelial cells by anti-inflammatory cytokines, this is not observed in hepatocytes (Liu et al, 2011).…”

“…These findings differed from those of other studies in which a large amount of horseradish peroxidase ( MW = 40,000) diffused into the parenchyma in the OVLT (Herde et al, 2011 ) and ME (Broadwell et al, 1983 ; Rodríguez et al, 2010 ). Horseradish peroxidase is known to be incorporated by mannose receptor-mediated transcellular and clathrin-mediated transcytosis routes (Ellinger and Fuchs, 2010 ). In the medial basal hypothalamus, wheat germ agglutinin lectin ( MW = 38,000) was shown to be taken up by tanycytes in the arcuate nucleus (Arc) via cellular internalization (Peruzzo et al, 2004 ; Morita et al, 2013a ).…”
