Recombinant Interleukin-1 Triggers the Increase of Circulating Fibronectin Level in Rats

Abstract: Interleukin-1 (IL-1) is one of the mediators responsible for the acute phase protein response during inflammation. We evaluated the effect of recombinant IL-1α given intra-abdominally on rat circulating fibronectin (Fn) level. Circulating Fn level, sampled between 3 h and 7 days after injection, was maximum at 24–48 h in Wistar rats treated with IL-1 at doses of 10 U and 100 U/100 g body weight (BW), respectively. Circulating Fn level in rats injected with 100 U/100 g BW IL-1 was higher than that of rats given… Show more

“…We have observed that during the process of monocyte transmigration into the subendothelial space of the coculture the cells acquire the typical appearance of spread monocytes suggesting the activation of these cells (unpublished observations, and reference 61). Injection of IL-1 into rats has been reported to produce elevated serum levels offibronectin (62). Leukocyte interleukins have also been reported to induce cultured EC to produce a highly organized pericellular matrix (63).…”

“…30 Fibronectin has been shown to be involved in inflammation. In animals, it is a positive acute-phase protein, with higher blood levels during inflammation, 31 but in humans, plasma FN is regarded as a negative acute-phase protein. 32 Fibronectin promotes chemotaxis and influences leukocyte function.…”

“…More recently, IL-1p has been implicated in fever regulation through a direct action of the hypothalamus thermoregulation center and the stimulation of acute-phase protein synthesis by the liver (di Giovine and Duff, 1990). In addition, several recent studies have provided evidence that this cytokine may be an important regulator of connective tissue synthesis (Bitterman et al, 1986;Dedhar, 1989b;Duncan and Berman, 1989;Goldring and Krane, 1987;Hagiwara et al, 1989Hagiwara et al, , 1990. IL-1p stimulates proliferation of fibroblasts (Bitterman et al, 1986), affects collagen and fibronectin synthesis (Duncan and Berman, 1989;Goldring and Krane, 19871, modulates bone metabolism (Felix b;let al, 1989;Garrett and Mundy, 1989;Lorenzo et al, 1988), and induces chondrolysis (Saklatvala et al, 1985).…”
