Recombinant Protein Production of Earthworm Lumbrokinase for Potential Antithrombotic Application

Abstract: Earthworms have been used as a traditional medicine in China, Japan, and other Far East countries for thousands of years. Oral administration of dry earthworm powder is considered as a potent and effective supplement for supporting healthy blood circulation. Lumbrokinases are a group of enzymes that were isolated and purified from different species of earthworms. These enzymes are recognized as fibrinolytic agents that can be used to treat various conditions associated with thrombosis. Many lumbrokinase (LK) g… Show more

“…LK is a fibrinolytic enzyme originated from earthworm which has shown to possess antithrombotic action (Wang et al 2013). The micelles consisted of a block copolymer of polycaprolactone-b-poly(2-(dimethylamino) ethyl methacrylate) (PCL-PDMAEMA), as well as a block copolymer of methoxy polyethylene glycol-b-polycaprolactone (mPEG-PCL) missed with a block copolymer of polycaprolactone-b-polyethylene glycol with an RGD peptide conjugated (PCL-PEG-RGDfk) forming a treatment known as LKTM (Pan et al 2018).…”

“…The molecular weights of LK proteins are in the range of 20 to 40 kDa and their isoelectric points (pI) are about 3 to 5. Since prokaryotic cells are not capable of performing post‐translational modifications, Escherichia coli was not suggested to express eukaryotic LK proteins with proper folding, processing, and glycosylation . Studies have shown that LK proteins contain glycan chains .…”

“…There is an increasing demand for LK protein for the development of various applications of LK in the treatment of thrombotic and vascular disease therapies. LK is also considered a very good candidate for recombinant production in a plant expression system . Unlike most proteins, LK proteins can be absorbed in the gastrointestinal tract intact and retain activity .…”
