“…This observation suggests that PKC-α is most likely to be involved in the inhibitory action of TPA on Ca 2+ signalling through the CCK receptor. The same conclusion was recently reached in studies of CHO-K1 cells [17] and Willems, unpublished observations) that stably express the rat pancreatic acinar CCK receptor. Importantly, CHO-09 cells do not express PKC-ε which makes it less likely that this isotype has a role.…”

“…Proteins were transferred overnight to polyvinylidine difluoride membranes (Immobilon P, Millipore) by Western blotting. Membranes were blocked for 1 h with PBS (pH 7.4) containing 0.2% (w/v) I-Block reagent and 0.1% (v/v) Tween-20 and incubated overnight with PKC-isotype-specific antibodies diluted 1:500 in PBS containing 0.1% (w/v) I-Block reagent and 0.2% (v/v) Tween-20 as described previously [17]. To demonstrate the specificity of the reaction, control membranes were incubated with PKC-isotype-specific antibodies in the presence of corresponding PKC-isotype-specific peptides diluted 1:1000.…”

“…We have previously shown that activation of PKC by the phorbol ester 12‐ O ‐tetradecanoylphorbol 13‐acetate (TPA) inhibits high‐affinity Ca 2+ signalling through the CCK A receptor in freshly isolated pancreatic acinar cells (Willems et al , 1993b; 1995) and stably transfected CHO‐K1 cells (Smeets et al , 1996). The aim of the present work was to compare modes of signal transduction and regulation of receptor functioning by PCK of pure CCK A and CCK B receptors after functional expression in the same eukaryotic expression system, namely the CHO‐09 cell.…”

“…Western blotting was performed essentially as described previously [27]. Briefly, total cell lysates from CHO-CCK A cells and rat pancreatic acini and the membrane and cytosol fractions prepared from both cell types were subjected to SDS-PAGE.…”
