Recruited Macrophages Control Dissemination of Group A Streptococcus from Infected Soft Tissues

Abstract: Group A Streptococcus (GAS) causes diverse infections in humans, ranging from mild to life-threatening invasive diseases, such as necrotizing fasciitis (NF), a rapidly progressing deep tissue infection. Despite prompt treatments, NF remains a significant cause of morbidity and mortality, even in previously healthy individuals. The early recruitment of leukocytes is crucial to the outcome of NF; however, although the role of polymorphonuclear neutrophils (PMNs) in host defense against NF is well established, th… Show more

“…A novel finding of this study is that neutrophils are required for the selection of CovRS mutations. Recruited macrophages play a role in clearance of GAS (43). However, macrophages appear not to play a critical role in the selection of covRS mutations in MGAS2221, since the decrease in the selection of SpeB AϪ variants of MGAS2221 in CXCR2 Ϫ/Ϫ mice is correlated with the impaired neutrophil recruitment but not with the increased levels of macrophages.…”

“…In this model, animals experience spreading necrosis, followed by bacteremia and mortality closely resembling severe necrotizing fasciitis in humans (21,22). We investigated TTP function in this infection model by using TTP ΔM mice, which lack TTP in neutrophils, monocytes, and macrophages (12), all of which are essential for defense against S. pyogenes infection (23)(24)(25)(26). Infection of Ttp fl/fl control animals (referred to herein as TTP fl/fl mice) led to 90% lethality (humane endpoint) 5 days post infection (p.i.…”
