RED AND FAR-RED INSENSITIVE 2, a RING-Domain Zinc Finger Protein, Mediates Phytochrome-Controlled Seedling Deetiolation Responses

Abstract: Light is arguably the most important resource for plants, and an array of photosensory pigments enables plants to develop optimally in a broad range of ambient-light conditions. The red-and far-red-light-absorbing photosensory pigments or phytochromes (phy) regulate seedling deetiolation responses, photoperiodic flowering, and circadian rhythm. We have identified a long hypocotyl mutant under red and far-red light, rfi2-1 (red and far-red insensitive 2 to 1). rfi2-1 was also impaired in phytochrome-mediated en… Show more

“…Based on these interaction screens 5 RING E3 ligases were selected to the study, represented by the following mutant lines; N656705 (AT5G65683), N686069 (AT1G61620), N372291 (AT3G29270), N2037522 and N67002 (AT4G17680), and N742646 (AT2G44410). In addition, a mutant line for COP1, cop1-6 , and RED AND FAR-RED INSENSITIVE 2 (RFI2) for which a role in mediating red and far-red light signaling and ubiquitination activity has been shown in vitro , were included (Stone et al, 2005; Chen and Ni, 2006a). This E3 ligase was selected as a candidate since its expression is regulated by circadian clock and rfi2-1 mutant flowers early (Chen and Ni, 2006b).…”

“…By reverse genetics, we have identified candidate genes to regulators of shade-avoidance responses; ATHB2 orthologs (Ciarbelli et al 2008), photomorphogenesis and seedling establishment; FHY3/FAR1, NDPK2, PIF/PIL family, PP7, RAP1/ ATMYC2, RFI2 (Shen et al 2005;Yadav et al 2005;Chen and Ni 2006;Lin et al 2007;Genoud et al 2008;Leivar et al 2008), light-regulated proteolysis; COP1, EID1, LAF1, SPA1 (Yi and Deng 2005;Marrocco et al 2006;Yang et al 2009;Chen et al 2010;Fankhauser and Ulm 2011) and developmental transitions; CO, PFT1 (SuĂĄrez-LĂłpez et al 2001;CerdĂĄn and Chory 2003) (Table S2). Using the obtained sequence information in a reverse genetics approach, we have identified a novel petunia mutant carrying a mutation in CCT (CO, CO-like, TOC1) domain (Strayer et al 2000;Valverde 2011).…”

“…In addition, the homologous PHYB transcript of N. attenuata was significantly down‐regulated in both ir‐ dcl3 and ir‐ dcl4 lines (Table S1). Light receptors entrain endogenous diurnal rhythms in plants and also control the plant's sensitivity to jasmonates during herbivore attack in Arabidopsis (Chen and Ni 2006). Interestingly, herbivores perform better on plants exposed to far‐red light, and total phenolics are lower in phyb mutants than in wild type plants (Moreno et al 2009).…”
