Red blood cells under varying extracellular tonicity conditions: an optical tweezers combined with micro-Raman study

Abstract: Extracellular tonicity has a significant influence on human red blood cell deformation capability. Advancements in the area of laser physics and optical trapping have opened up a plethora of applications for understanding cell structure and dynamics. Here, Raman Tweezers technique was employed to investigate the impact of extracellular tonicity by exposing human red blood cells to both hypertonic and hypotonic intravenous fluids. Heme aggregation was observed in hypertonic saline solution, accompanied with dam… Show more

“…Raman peaks that displayed enhanced intensity in this spectral region were wavenumbers at 1,212, 1,246, and 1,264 cm –1 . Alteration of 1,246 cm –1 is indicative of heme aggregation ( 41 ). This was accompanied by a fall in intensity at the peak of 1,222 cm –1 suggesting changes in the methine C-H deformation ( 35 , 36 ).…”

“…A new peak surfaced at 663 cm –1 that shouldered the 675 cm –1 pyrrole-deformation peak across the disease populations as well. Both these events are indicative of heme-aggregation in consequence of protein denaturation ( 40 , 41 ). The porphyrin breathing mode peak at 752 cm –1 , signifying Hb integrity ( 45 ), exhibited a significantly reduced intensity in the disease groups.…”

“…The weak signals in Raman scattering can be enhanced by placing the sample in contact with a metallic nanoparticle surface / roughened metallic film surface in order to satisfy plasmonic condition, which is termed as Surface Enhanced Raman Spectroscopy. The Raman Tweezers technology combines optical tweezers with Raman spectroscopy, where the micron sized particle in aqueous solution will be trapped using a laser beam and will be probed/characterized using Raman spectroscopy [9][10][11]. The FTIR technique is used to capture the infrared spectrum of absorption of substances that are solid, liquid, or gaseous.…”

“…Tonicity is an essential factor for cells, which affects the metabolic activity and the exchange of substances with the extracellular region [1,2]. Changes in tonicity affect transmembrane ion transport by activating or inactivating specific ion channels [3], which is the characteristic of certain cells with tonicity imbalance due to disease [4].…”
