Red human hair pheomelanin is a potent pro‐oxidant mediating UV‐independent contributory mechanisms of melanomagenesis

Abstract: The highest incidence of melanoma in red haired individuals is attributed to the synthesis and phototoxic properties of pheomelanin pigments. Recently, pheomelanin has also been implicated in UV-independent pathways of oxidative stress; however, the underlying mechanisms have remained uncharted. Herein, we disclose the unprecedented property of purified red human hair pheomelanin (RHP) to promote (i) the oxygen-dependent depletion of major cell antioxidants, for example glutathione and NADH; (ii) the autoxidat… Show more

“…[ 36,42 ] The impedance vs. frequency (Z vs. f) spectra readouts and subsequent extraction of the real (ε′) and imaginary (ε′′) part of permittivity and loss factor tan δ = ε′/ε′′ are sensitive methods to discriminate if a medium behaves like a biological or as a (poly)electrolyte material, because of the well known typical features of the impedance spectra of biological media (see S.I. and Figure S3).…”

“…Recently, pheomelanin structure has been elucidated and pheomelanin was purified [22, 23]. The purified pheomelanin exhibited potent pro-oxidant characteristics in the test tube and in cells when exposed to UV radiation [24, 25]. Further investigations should lead to novel mechanistic insights about UV-induced melanomagenesis.…”

“…The recent studies of pheomelanin have also proved that it is involved in UVindependent pathways of oxidative stress. [12][13][14] Therefore, it is important to find effective antioxidant which will neutralize produced harmful free radicals, especially those produced by pheomelanin.…”
