Redox characteristics of normal and habituated cell lines of sugarbeet

Abstract: Redox properties of cells of normal hormone-requiring (N) and habituated (H) sugarbeet calli have been investigated. It was found that H cells at all ages reduced exogenous ferricyanide at a much higher rate than N cells, but they exhibited a lower chemiluminescence either alone or in presence of luminol. The plasma membrane NADH:ferricyanide oxidoreductase measured in vitro was almost identical for the two cell types. This could indicate that the higher reducing power of H cells had an intracellular origin. A… Show more

“…An experimental system composed of in vitro sugar beet callus lines originating from the same mother plant but exhibiting different status of differentiation was used (Cerrie et al 1994, Hagège 1996, Causevic et al 2005). Distinct treatments inducing DNA hypo‐ or hypermethylation have already been shown to generate variations of morphogenetic status: loss of organogenic potential and acquisition of dedifferentiation.…”

“…Thus, the developmental fate of regenerating tissue in culture could be directed by the plant hormones cytokinin and auxin that act on chromatin structure and particularly on DNA methylation (Zhao et al 2008). Furthermore, O and NO lines have been generated several years ago and used for several studies (Cerrie et al 1994, Hagège 1996, Causevic et al 2005, 2006), while the other lines were generated for the present study. The effect of aging on DNA methylation has already been reported but with contradictory trends (Fraga et al 2002, Sha et al 2005, Valledor et al 2007).…”

“…In contrast, prolonged exposure to high ROS doses, as in the DD line, will induce cell damage and/or proliferation. Results dealing with cell redox state have been reported more than 10 years ago for the sugarbeet DD and NO lines only (Hagège et al 1992;Cerrie et al 1994;Cherif et al 1996;Hagège 1996).…”

“…Recently, we have demonstrated a relationship between phenotypes of these cell lines, their ligniWcation during cell diVerentiation and their DNA methylation levels (Causevic et al 2005). Previous reports on cellular redox state parameters have been published in other independent works only for the DD and NO lines and have revealed an oxidative stress state in the DD line (Hagège et al 1992;Cerrie et al 1994;Cherif et al 1996;Hagège 1996). These cell lines represent an interesting model for the study of plant morphogenesis; and reactive oxygen species (ROS) are believed to play an important role as intracellular messengers during morphogenesis (Sauer et al 2001;Boonstra and Post 2004;Mittler et al 2004;Edreva 2005).…”
