Redox Control of the Senescence Regulator Interleukin-1α and the Secretory Phenotype

Abstract: Background:The senescent microenvironment is permissive to disease progression, and the role of oxidants in this process remains uncharacterized. Results: Senescent fibroblasts promote tumor invasion through redox/calcium regulation of the cytokine IL-1␣. Conclusion: Senescence-associated oxidants and calcium drive the secretory phenotype, altering the microenvironment. Significance: Targeting senescent cells with antioxidant-based therapeutics may restrict inflammation and combat age-related disease progressi… Show more

“…Our observations suggest that senescent disc cells also prefer to 82 aggregate in monolayer cultures (Fig.3). “…The following examples further illustrate the use of kinetically controlled systems in nuclear protein trafficking and in maintenance of the integrity of the genome. Recent reviews are also available on the redox processing in the secretory pathway [33, 34, 120] and in mitochondrial import [35, 36]. Relatively less information is available on vectorial redox processing during endocytosis and proteolysis in lysosomes, but evidence is available suggesting that analogous systems are present [121, 122].…”

“…Apoptotic endothelial cells contain the full-length IL-1a precursor (70); IL-1alpha appears to be a DNA damage sensor (71) and play a role in cellular senescence (72). Whether IL-1 alpha plays an important role in inflammation and autoimmunity in PAH remains to be investigated.…”
