Reduced Expression of the Interferon‐Gamma Messenger RNA in IgG2 Deficiency

Abstract: The specific defect that causes IgG2 deficiency, which is one of the primary immunodeficiencies, is unknown. Recently, it was shown that interferon‐γ (IFN‐γ) induces synthesis of human germline Cγ2 transcripts. In the authors’ previous study and the present one, peripheral blood lymphocytes (PBLs) of all five tested patients with IgG2 deficiency failed to produce enough IFN‐γ when stimulated with phytohaemagglutinin or concanavalin A although they produced a sufficient amount of interleukin‐2 (IL‐2). The low l… Show more

“…The cytokine regulation of IgG2 production is still a largely ambiguous subject. Gamma interferon (IFN-␥) may have a role, as suggested in previous studies (9,12,14). However, we were unable to note any significant difference in IFN-␥ level between the mild and severe groups in our study.…”

“…No relationship between anti‐ P. falciparum IgE and IgG1, and/or IgG3 levels was observed. IgG2 is known to be a Th1 subclass Ig‐dependent on IFN‐γ (29,30). According to recent evidences, IL4 significantly enhances the expression of ɛ, γ1, γ2, γ3 and γ4 germline gene transcript (GLT) and the secretion of the corresponding proteins.…”

“…Specific cytokine responses in the SPT − subjects involves IFNγ, IL‐6 and IL‐10 production at levels comparable to the SPT + group, together with very low levels of IL‐4 mRNA. It is pertinent to note in this context that IgG2 antibody production has been shown to be IFNγ dependent [ 24, 25].…”
