PurposeTo establish the relationship between plasma levels of thioredoxin (Trx) and macrophage migration inhibitory factor (MIF) in systemic inflammatory stress syndrome (SIRS)/sepsis.MethodsEnzyme-linked immunosorbent assay measurements of Trx, MIF, IL-6, -8, and -10 and enzyme-linked fluorescent assay determination of procalcitonin (PCT) in plasma from patients with SIRS/sepsis, neutropenic sepsis, healthy volunteers and pre-oesophagectomy patients.ResultsThioredoxin was significantly higher in SIRS/sepsis patients [101.3 ng ml−1, interquartile range (IQR) 68.7–155.6, n = 32] compared with that in healthy controls (49.5 ng ml−1, IQR 31.4–71.1, P < 0.001, n = 17) or pre-oesophagectomy patients (40.5 ng ml−1, IQR 36.9–63.2, P < 0.01, n = 7), but was not raised in neutropenics (n = 5). MIF levels were also significantly higher in SIRS/sepsis patients (12.1 ng ml−1, IQR 9.5–15.5, n = 35), but not in the neutropenic group, when compared with healthy controls (9.3 ng ml−1, IQR 7.3–10.7, P < 0.01, n = 20). Trx levels correlated, positively, with MIF levels and APACHE II scores. Plasma levels of IL-6, -8 and -10 and PCT increased significantly in patients with SIRS/sepsis (P < 0.001) and with neutropenic sepsis, but did not correlate with Trx or MIF levels.ConclusionPlasma levels of Trx, MIF, IL-6, -8, -10 and PCT were raised in patients with SIRS/sepsis. Comparisons between mediators suggest a unique correlation of Trx with MIF. Moreover, Trx and MIF differed from cytokines and PCT in that levels were significantly lower in patients with neutropenia compared with the main SIRS/sepsis group. By contrast, IL-8 and PCT levels were significantly greater in the neutropenic patient group. The link between MIF and Trx highlighted in this study has implications for future investigations into the pathogenesis of SIRS/sepsis.

The AIM of this experimental study was to evaluate the effect of the antioxidant drug "U-74389G" in a rat model of hypoxia reoxygenation (HR) using the previously established protocol. Effects of treatment were evaluated by mean red blood cell distribution width (RDW) levels. MATERIALS AND METHODS: 40 rats of a mean weight of 231.875 g were employed in the study. RDW levels were determined at 60 min (groups A and C) and at 120 min (groups B and D) after starting the reoxygenation. Groups A and B received no drugs, whereas rats from groups C and D were administered with U-74389G. RESULTS: demonstrated that U-74389G administration signifi cantly decreased the RDW levels by 4.96% + 2.27% (p = 0.0175). Reoxygenation time non-signifi cantly decreased the RDW levels by 0.27% + 2.41% (p = 0.8889). Together, U-74389G administration and reoxygenation time nonsignifi cantly decreased the RDW levels by 2.54% + 1.39% (p = 0.0679). CONCLUSIONS: U-74389G administration particulary in concert without reperfusion declines the RDW levels even within the short -time context of 1.5 hours reperfusion. Эффективность лечения была оценена путём измерения среднего отклонения распределения эритроцитов (ОРЭ). МАТЕРИАЛЫ И МЕТОДЫ: Исследование проводилось на 40 крысах средним весом 231.875 g. Среднее распределение эритроцитов было измерено через 60 минут (группы А и В) и через 120 минут (группы Б и Г) после начала реоксигенации. Лекарственный продукт U-74389G ввели только животным из групп В и Г, в группах А и Б его не применяли. РЕЗУЛЬТАТЫ: Результаты показывают, что U-74389G сигнификантно понижает ОРЭ на 4.96% + 2.27% (p = 0.0175). Время реоксигенации понижает ОРЭ несигнификантно на 0.27% + 2.41% (p = 0.8889). В комбинации U-74389G и реоксигенация понижают ОРЭ несигнификантно на 2.54% + 1.39% (p = 0.0679). ЗАКЛЮЧЕНИЕ: Применение U-74389G и особенно его взаимодействие с временем для реперфузии понижает ОРЭ, даже и за время короткой реперфузии в рамках 1.5 часа. Abstract: Administration of U-74389G, which has radical scavenging properties, attenuates leukocyte adherence in selected populations of intestinal venules which is found increased during endotoxemia. Thus, 21-aminosteroids may have an impact in the treatment of endotoxin-induced intestinal injury.
