Reduction of Cisplatin Ototoxicity by Fosfomycin in Animal Model

Abstract: The protective effect of fosfomycin against cisplatin-induced ototoxicity was studied in rats. Sixty-four Fischer rats were injected intravenously with daily doses of 1, 2, 5, and 10 mg/kg of cisplatin with or without 300 mg/kg of fosfomycin for a varying period from 1 to 10 days. The total dose of 10 mg/kg of cisplatin was given equally in all animals. Inner ear damage appeared to be more reduced histopathologically in animals given both cisplatin and fosfomycin than in animals given cisplatin alone. Similarl… Show more

“…Higher doses of cisplatin in the rat have been associated with significant mortality. [24][25][26] Three milliliters of sterile 0.9 percent saline were administered immediately after each cisplatin injection to counteract the nephrotoxicity of the drug. 22,24 Rats received rat chow and water ad libitum and were housed four to a plastic cage that contained wood chips.…”

“…There are also many reports of the protective action of fosfomycin against drug-induced nephrotoxicity (Inouye and others 1982, Fujita and others 1983, Ohtani and others 1984, Yogi and others 1989, Shibuya and others 1990, Wada and others 1995, Gobernado 2003). However, there have been no reports of nephrotoxicity associated with fosfomycin administration in human beings or animals.…”
