Region-specific expression of nitric oxide synthases in the bovine oviduct during the oestrous cycle and in vitro

Abstract: Nitric oxide synthases (NOS) account for the endogenous production of nitric oxide (NO), a small and permeable bioreactive molecule. NO is known to act as a paracrine mediator during various processes associated with female reproduction. In the present study, the mRNA expression of the endothelial (eNOS) and inducible (iNOS) NO synthases were examined in bovine oviduct epithelial cells (BOEC) during the oestrous cycle. In addition, eNOS and iNOS mRNA and protein were localised by in situ hybridisation and immu… Show more

“…6). In addition, partial differences of NO synthases in cyclic epithelium of the oviduct (Ulbrich et al 2006) and whole layers containing stroma and smooth muscle of the oviduct (Fig. 1) suggest layer-specific control of NO synthesis in the oviductal milieu.…”

“…The abundance of mRNA for the ribosomal protein S18 (S18) and for the Na + /K + -ATPase α subunit was quantified by qRT-PCR using the iCycler and the iQ-SYBR green supermix (Bio-Rad Laboratories GmbH, Munich, Germany) as described previously Ulbrich et al, 2006). Briefly, 1 μl aliquots of each RT reaction (1/20 of total) were primed, in each 10 µl PCR, with gene-specific oligonucleotides (final concentration of 0.2µM) for S18 (gi 74268022: 5'-CTT AAA CAG ACA GAA GGA CGT GAA-3' and 5'-CCA CAC ATT ATT TCT TCT TGG ACA-3'; transcript size: 218 bp) and Na + /K + -ATPase α (gi 115305283: 5'-GAG ATT ACC CCC TTC CTG ATA TTT-3' and 5'-TGG ATC ATA CCA ATC TGT CCA TAG-3'; transcript size: 227 bp).…”
