Leaf rust caused by the fungus Puccinia triticina is one of the most important diseases of wheat (Triticum aestivum) worldwide. The use of resistant wheat cultivars is considered the most economical and environment-friendly approach in controlling the disease. The Lr38 gene, introgressed from Agropyron intermedium, confers a stable seedling and adult plant resistance against multiple isolates tested in Europe. In the present study, 94 F 2 plants resulting from a cross made between the resistant Thatcherderived near-isogenic line (NIL) RL6097, and the susceptible Ethiopian wheat cultivar Kubsa were used to map the Thatcher Lr38 locus in wheat using simple sequence repeat (SSR) markers. Out of 54 markers tested, 15 SSRs were polymorphic between the two parents and subsequently genotyped in the population. The P. triticina isolate DZ7-24 (race FGJTJ), discriminating Lr38 resistant and susceptible plants, was used to inoculate seedlings of the two parents and the segregating population. The SSR markers Xwmc773 and Xbarc273 Xanked the Lr38 locus at a distance of 6.1 and 7.9 cM, respectively, to the proximal end of wheat chromosome arm 6DL. The SSR markers Xcfd5 and Xcfd60 both Xanked the locus at a distance of 22.1 cM to the distal end of 6DL. In future, these SSR markers can be used by wheat breeders and pathologists for marker assisted selection (MAS) of Lr38-mediated leaf rust resistance in wheat.

Background Many times small differences in genome size are reported between or within plant species in which no cytologic confirmation is made. Attempts to repeat these studies have met with limited success. The controversy then becomes whether or not these small differences that were not confirmed cytologically are real. The present study was undertaken to determine if the ≈1% nuclear variation seen by flow cytometry among wheat lines selected for aluminum response was due to actual chromatin differences. Methods The three parental wheat cultivars used in the aluminum selection along with the isolines resulting from the selection were analyzed. One parental line had previously been reported to have alien chromatin substituted for the corresponding wheat homoeologous chromatin. Genomic in situ hybridization was used to determine the presence or absence of rye chromatin in three cultivars and six near‐isolines of wheat. Results Upon observing metaphase chromosomes of the Century parent and its isolines, two of the chromosomes were observed to be one‐half yellow‐orange, indicating rye chromatin with the remaining portion of the chromosomes and the other 40 wheat chromosomes having no label indicating wheat chromatin. In the Chisholm parent and its isolines, none of the chromosomes were labeled, indicating the absence of rye chromatin. In addition, none of the third parents' chromosomes had the rye yellow‐orange signal. Conclusions The wheat lines with the larger DNA contents were observed to have alien DNA present. DNA differences between the normal wheat chromosomes and the substituting alien chromatin were calculated based on total chromosome length. The increase in genome size of the wheat lines containing the alien chromatin appears to be the result of the alien chromatin having ≈43% more DNA than the wheat chromatin it is replacing. Thus, the small DNA difference previously reported by flow cytometry was demonstrated to be a real DNA variation due to the presence of small fragments of alien chromosomes added to the wheat genome. Endopeptidase allele Ep-D1c and DNA marker-assisted selection have been used for the incorporation of Lr19 + Lr24 leaf rust resistance genes combination into adapted commercial winter wheat cultivars. The first step was the transfer of the gene Lr19 from the donor cultivar Agrus into acceptor cultivars Simona and Lívia. The progenies possessing the null allele Ep-D1c linked to the gene Lr19 have been screened for their resistance to leaf rust by isolate 4332 SaBa. The plants homozygous properties at the Ep-D1c locus and resistant against leaf rust were used for crossing with NIL Thatcher/Lr24 -a donor of the gene Lr24. Plants possessing both Lr genes were selected from F 2 population by STS and isozyme markers linked to the Lr genes. Progenies of 18 F 2 plants have been selected by STS marker and tested for resistance against leaf rust. Results obtained with isozyme and STS markers corresponded with resistance testing. Altogether 6 progenies of F 3 generation possessing a resistance gene combination of Lr19 + Lr24 in a homozygous condition were developed.
