Regulation of a Trehalose-Specific Facilitated Transporter (TRET) by Insulin and Adipokinetic Hormone in Rhodnius prolixus, a Vector of Chagas Disease

Abstract: Using the blood-sucking kissing bug, Rhodnius prolixus as an experimental model, we have studied the involvement of insulin-like peptides (ILPs) and adipokinetic hormone (AKH) signaling in carbohydrate metabolism, focusing on the regulation of the trehalose-specific facilitated transporter (Rhopr-TRET), particularly in the ovaries. We find that trehalose stores in ovaries increase after feeding, synchronously with the beginning of vitellogenesis, but that the transcript expression of enzymes involved in trehal… Show more

“…It is well known that reproductive processes are energetically demanding, so it is therefore no surprise that they undergo complex pathways related to lipid and carbohydrate availability, reallocation, and metabolism ( Fullbright et al, 1997 ; Nässel and Vanden Broeck, 2016 ; Leyria et al, 2021a ). It was shown that ILPs and ILP signalling are necessary for correct functioning of both the female and male reproductive systems.…”

“…The ligand adipokinetic hormone (AKH) synthesized and released by the corpora cardiaca (CC) and AKH receptor (AKHR) as rhodopsin‐like GPCR has been proven to be involved in the regulation of energy metabolism, feeding, reproduction, diapause, flight and other activities. Particularly under stress conditions, AKH and AKHR can couple and participate in antioxidant stress responses by activating AC, PKA and cAMP 37–45 . Recent studies have shown that AKH and its receptor AKHR are involved in regulating resistance to insecticides 38,46–49 .…”

[37][38][39][40][41][42][43][44][45] Recent studies have shown that AKH and its receptor AKHR are involved in regulating resistance to insecticides. 38,[46][47][48][49] The AKH titer in hemolymph of the locust Schistocerca gregaria increased post-treatment with deltamethrin.…”

“…A previous study demonstrated that higher bombyxin signaling was detected in HCl-treated eggs compared to diapause eggs (Gu et al, 2019). Future that plays an important role in regulating trehalose homeostasis (Kikawada et al, 2007;Leyria et al, 2021). Transcriptional regulation of theTret gene by the juvenile hormone was reported in T.…”

“…Facilitation of trehalose transporter 1 (Tret1), a conserved transporter in insects, plays a role in maintaining hemolymph trehalose levels in insects (Kanamori et al, 2010; Kikawada et al, 2007). Insulin signaling appears to positively affect Tret1 transcript expression in the ovaries of fed Rhodnius , a state where anabolic processes are required for egg formation and where it is known that an insulin signaling pathway is activated (Leyria et al, 2021). In Drosophila , stage‐dependent upregulation of Tret1 expression by ecdysteroid signaling was reported (Nishimura, 2020).…”
