Regulation of c-jun gene expression in human T lymphocytes

Abstract: The present studies have examined the effects of mitogens on induction of early response gene expression in normal peripheral blood T and Jurkat cells. Pokeweed mitogen (PWM) or anti-CD3 significantly increases c-jun messenger RNA (mRNA) levels in T cells. This transient PWM-related increase in c-jun transcripts is maximal after 15 to 30 minutes of exposure of T cells to PWM. PWM induces c-jun gene expression in a concentration-dependent manner. Moreover, PWM similarly induces expression of other genes coding … Show more

“…This induction of the c-jun gene is similar to levels reported by other investigators [Angel et al, 19881. The increase in c-jun mRNA levels is due to an increased transcription rate [Quantin and Breathnach, 1988;Ryder and Nathans, 1988;Wu et al, 1989;Chauhan et al, 1993;Wang and Scott, 19941.…”

“…Members of the AP-1 family can also crossdimerize with ATF/CREB transcription factors [Hai and Curran, 19911. The c-jun gene is expressed in response to a variety of external stimuli, including growth factors, W radiation, ionizing radiation, oxidative stress, and various chemical agents [Sherman et al, 1990;Devary et al, 1991;Bergelson et al, 19941. Elevation of c-jun mRNA levels in response to these stimuli is a consequence of increased transcription of the gene [Quantin and Breathnach, 1988;Ryder and Nathans, 1988;Wu et al, 1989;Sherman et al, 1990;Devary et al, 1991;Chauhan et al, 1993;Bergelson et al, 19941. We have recently analyzed the promoter of the c-jun gene in HeLa cells during U V irradiation, which induces the gene > 100fold [Devary et al, 1991;Rozek and Pfeifer, 19931. Six transcriptional activator proteins were detected along the c j u n promoter region in HeLa cells. Surprisingly, no changes in protein-DNA interactions were observed during the UV response, indicating that induction of the gene was mediated by preformed protein-DNA complexes at the promoter.…”

“…AP-1-mediated activation of the HTLV-1 LTR resulted from direct interactions formed between AP-1 and TRE-1 repeat II [51,53]. AP-1 expression and DNA-binding activity have been shown to be up-regulated during monocytic differentiation and T cell activation by stimulation of the protein kinase C (PKC) pathway [40,55]. It is interesting that PKC-mediated activation of HTLV-1 gene expression has been shown to be dependent on TRE-1 repeat II [56].…”

“…c-Jun is the best characterized AP-1 family member that positively regulates cells proliferation (101) and is frequently overexpressed in several cancers including lymphomas (102)(103)(104). Stimulation of the antigen receptor or mitogen treatment significantly induces c-Jun gene expression in normal PBLs and Jurkat T cells (105). Interestingly, c-Jun is further regulated at the transcription level by its own gene product through a positive feedback loop (106).…”
